Development and Validation of a LC-ESI-MS/MS Method for the Determination of Alternaria Toxins Alternariol, Alternariol Methyl-Ether and Tentoxin in …

2016

Alternaria species are capable of producing several secondary toxic metabolites in infected plants and in agricultural commodities, which play important roles in food safety. Alternaria alternata turn out to be the most frequent fungal species invading tomatoes. Alternariol (AOH), alternariol monomethyl ether (AME), and tentoxin (TEN) are some of the main Alternaria mycotoxins that can be found as contaminants in food. In this work, an analytical method based on liquid chromatography (LC) tandem mass spectrometry (MS/MS) detection for the simultaneous quantification of AOH, AME, and TEN in tomato and tomato-based products was developed. Mycotoxin analysis was performed by dispersive liquid-…

Seeds vitality and fungal contamination in Abies nebrodensis

2022

Seeds of Abies nebrodensis were subjected to laboratory tests aimed to detect fungal contaminants and to obtain xenobiotic-free seedlings, by the use of different surface sterilising agents. Moreover, hot water at 60°C was used to suppress any fungal microorganisms colonizing the inner tissues. Alternaria alternata, Aspergillus flavus and Stemphylium vesicarium were the most frequent fungal contaminants. Non-contaminated seeds showed germination values ranging from 0 to 36.4% depending on the applied sterilization protocol. Further analyses will be carried out to establish the influence of these fungi on the seed germination process and their relationship with seedlings of A. nebrodensis.

A temperature-type model for describing the relationship between fungal growth and water activity

2001

Growth of Penicillium chrysogenum, Aspergillus flavus, Cladosporium cladosporioides and Alternaria alternata at their respective optimum temperatures was studied in Potato Dextrose Agar (PDA) medium at different water activities (a(w)) adjusted with glycerol. The growth rate (mu) was expressed as the increase in colony radius per unit of time. This paper extends the model that showed the relationship between temperature and bacterial growth rate developed by Rosso et al. [J. Theor. Biol. 162 (1993) 447] to describe the influence of a(w) on fungal development. An excellent correlation between the experimental data and the model predictions was obtained, the regression coefficients (r2) were …

Determination of Fungi and Multi-Class Mycotoxins in Camelia sinensis and Herbal Teas and Dietary Exposure Assessment

2020

In this paper, a study of fungal and multi-mycotoxin contamination in 140 Camellia sinensis and 26 herbal teas marketed in Latvia is discussed. The analysis was performed using two-dimensional liquid chromatography with time-of-flight mass spectrometry (2D-LC-TOF-MS) and MALDI-TOF-MS. In total, 87% of the tea samples tested positive for 32 fungal species belonging to 17 genera, with the total enumeration of moulds ranging between 1.00 &times

Modificētus mālus saturošu materiālu antimikrobiālā iedarbība

2017

Ietekme tika novērota sešiem no divpadsmit māla pulvera paraugiem Alternaria a…

Alternaria Mycotoxins in Food and Feed: An Overview

2017

Alternariais one of the major mycotoxigenic fungal genera with more than 70 reported metabolites.Alternariamycotoxins showed notably toxicity, such as mutagenicity, carcinogenicity, induction of DNA strand break, sphingolipid metabolism disruption, or inhibition of enzymes activity and photophosphorylation. This review reports on the toxicity, stability, metabolism, current analytical methods, and prevalence ofAlternariamycotoxins in food and feed through the most recent published research. Half of the publications were focused on fruits, vegetables, and derived products—mainly tomato and apples—while cereals and cereal by-products represented 38%. The most studied compounds were alternario…

Isotope Fractionation of Selenium During Fungal Biomethylation by Alternaria alternata

2011

The natural abundance of stable Se isotopes may reflect sources and formation conditions of methylated Se. We aimed at (1) quantifying the degree of methylation of selenate [Se(VI)] and (hydro)selenite [Se(IV)] by the fungus Alternaria alternata at pH 4 and 7 and (2) determining the effects of these different Se sources and pH values on 82Se/76Se ratios (δ82/76Se) in methylselenides. Alternaria alternata was incubated with Se(VI) and Se(IV) in closed microcosms for 11-15 days and additionally with Se(IV) for 3-5 days at 30 °C. We determined Se concentrations and δ82/76Se values in source Se(VI) and Se(IV), media, fungi, and trapped methylselenides. In Se(VI) incubations, methylselenide vola…

Effect of analogues of plant growth regulators on in vitro growth of eukaryotic plant pathogens

2004

FGA (furfurylamine; 1,2,3,4 tetra-O-acetyl-β-d-glucose; adipic acid monoethyl ester), a chemical mixture of three analogues of plant growth regulators that increases the protection of tomato plants against phytopathogens, was demonstrated to have direct antimicrobial activity. It reduced the growth in vitro of the filamentous fungi Alternaria solani and Botrytis cinerea, and the oomycetes Phytophthora capsici and Phytophthora citrophthora (ED50 0·18–0·26% w/v, depending on species). The components of this mixture were also active against these phytopathogens, but sensitivity to the compounds was different for each pathogen. Adipic acid monoethyl ester (E) showed the highest and widest range…

Characterization of Alternaria alternata Isolates from Tangerine Hybrids Affected by Brown Spot in Italy

2011

Alternaria brown spot, caused by Alternaria alternata, is the most important disease of tangerines and their hybrids worldwide, including Italy. The disease represents, for some susceptible citrus cultivar, a limiting factor in profitable cultivation. The results of the characterization of A. alternata isolates obtained from lesions on leaves and fruits of Fortune, Nova and Winola hybrids and Valencia sweet orange, collected in two groves located in Calabria and Sicily, is reported. An endopolygalacturonase gene (endoPG) was sequenced from five representative strains and a phylogenetic tree based on endoPG sequence data of Italian isolates and those of citrus associated Alternaria species d…

Three novel synthetic amides of adipic acid protect Capsicum anuum plants against the necrotrophic pathogen Alternaria solani

2003

Three novel amides of adipic acid, 5-carbamoil ethyl pentanoate (N1), 5-(2-furfurylmethylcarbamoil) ethyl pentanoate (N2) and 5-(3-aminopropylcarbamoil) ethyl pentanoate (N3) were synthesized and their resistance-inducing activity was studied on pepper (Capsicum annuum) plants. Treatment with low doses of each amide protected pepper plants against the pathogen Alternaria solani, N1 being the most effective. Although their mode of action is still unknown, prechallenge studies demonstrated the induction of the phenylpropanoid pathway and an antisenescence effect. These effects were previously shown for other mixtures and derivatives of adipic acid. These new chemicals showed no antimicrobial …
