Azulene as a biphenyl mimetic in orexin/hypocretin receptor agonists

Azulene is a rare ring structure in drugs, and we investigated whether it could be used as a biphenyl mimetic in known orexin receptor agonist Nag 26, which is binding to both orexin receptors OX1 and OX2 with preference towards OX2. The most potent azulene-based compound was identified as an OX1 orexin receptor agonist (pEC50 = 5.79 +/- 0.07, maximum response = 81 +/- 8% (s.e.m. of five independent experiments) of the maximum response to orexin-A in Ca2+ elevation assay). However, the azulene ring and the biphenyl scaffold are not identical in their spatial shape and electron distribution, and their derivatives may adopt different binding modes in the binding site. New heteronuclear gold(I)-platinum(II) complexes with cytotoxic properties: are two metals better than one?

A series of mono- and heterodinuclear gold(I) and platinum(II) complexes with a new bipyridylamine-phosphine ligand have been synthesized and characterized. The X-ray structures of the ligand precursor 4-iodo-N,N-di(pyridin-2-yl)benzamide, and of one gold derivative are reported. All the complexes display antiproliferative properties in vitro in human cancer cells in the range of cisplatin or higher, which appear to correlate with compounds' uptake. Interestingly, studies of the interactions of the compounds with models of DNA indicate different mechanisms of actions with respect to cisplatin. Oxazoline-containing compatibilizers for polyamide/SAN and polyamide/ABS blends

Polyamide (PA) and acrylonitrile/butadiene/styrene copolymer (ABS) may appear as a mixture in the recycled plastic stream. The incompatibility of these blends results in a blend with poor mechanical properties. The aim of this work is to partially convert the nitrile groups of the acrylonitrile/styrene copolymer (SAN) into oxazoline groups by reaction with aminoethanol (AE). Such modified SAN (SAN-m) can react with the amine or carboxylic acid end groups of PA, and therefore used as compatibilizers for blends of PA with ABS. Dermatofibrosarcoma protuberans (DFSP) is a rare superficial tumor characterized by high rates of local recurrence and low risk of metastasis. DFSP occurs most commonly on the trunk and proximal extremities, affects all races, and often develops between the second and fifth decade of life. The tumor grows slowly, typically over years. Histologically, several variants of DFSP have been described and should be well characterized to avoid misdiagnosis with other tumors. Effects of carboxyamidotriazole on in vitro models of imatinib-resistant chronic myeloid leukemia.

Although imatinib mesylate (IM) has revolutionized the treatment of chronic myeloid leukemia (CML), some patients develop resistance with progression of leukemia. Alternative or additional targeting of signaling pathways deregulated in bcr-abl-driven CML cells may provide a feasible option for improving clinical response and overcoming resistance. Synthesis and antifungal activity of new N-isoxazolyl-2-iodobenzamides

N-Isoxazolyl-2-iodobenzamides 3 and 9, with a benodanil-like structure, were synthesized by refluxing in acetic acid the corresponding benzotriazinones 2 and 8 with potassium iodide for 1 h with the aim to ascertain if they were active as fungicides against Phytophthora citricola Saw., Botrytis cinerea Pers., Rhizoctonia sp. and Alternaria sp. Among the tested iodo derivatives, compounds 3b and 9a possess interesting activities against the aforesaid fungal strains in several cases similar to that of benodanil I taken as reference drug.

In this paper we present the synthesis and the characterization of cyclophosphazenes substituted with allyl groups, their transformation in epoxide-containing cyclophosphazenes and the final utilization of these compounds as chain extenders in combination with polyamides. The reaction at high temperature of Nylon-6 with epoxy-functionalized cyclophosphazenes leads to the opening of the epoxy units by the action of both amino (--NH2) and carboxylic (--COOH) end-groups of the polymer to enhance the final molecular weight of this material.
