Considerable interest is focused on overcoming multidrug resistance (MDR) in cancer chemotherapy. The in vitro experiments to characterize P-glycoprotein's (P-gp) function and to decrease its effects have led to a variety of strategies such as addition of competitors or supplementation of the medium with oligonucleotides complementary to the 5'-end of the MDR1-mRNA. For the Caco-2 cell line, an in vitro model for absorption screening, expressing multiple transporters including P-gp, which pumps substances back into the apical solution, P-gp activity might mask other relevant transport proteins' activity. Abstract The DNA-binding of two salphen-like metal complexes of nickel(II) (1) and zinc(II) (2) was investigated in different ionic strength solutions by absorption spectroscopy. The data analysis allowed us to obtain the values of their extrapolated DNA-binding constant in physiological conditions, with DNA-binding strength in the order Ni > Zn, and to give relative weight to the electrostatic and non-electrostatic contributions to their DNA-interaction.

Chemical initiation for butyl and lauryl acrylate monolithic columns for CEC.

Butyl acrylate (BA)- and lauryl acrylate (LA)-based monolithic stationary phases for CEC were synthesized, using a redox system as initiator of polymerization. BA monoliths were initiated with ammonium peroxodisulfate, whereas LA columns were obtained with lauroyl peroxide as initiator. In both cases, TEMED was used to activate the process. The influence of porogenic solvent composition on both morphological and electrochromatographic properties of the resulting monoliths was investigated. Excellent efficiencies (minimum plate heights of 4.2-6.3 microm for BA columns and 2.6-5.3 microm for LA stationary phases, for a PAHs mixture) were achieved. Label-Free Pyrophosphate Recognition with Functionalized Asymmetric Nanopores

[EN] The label¿free detection of pyrophosphate (PPi) anions with a nanofluidic sensing device based on asymmetric nanopores is demonstrated. The pore surface is functionalized with zinc complexes based on two di(2¿picolyl)amine [bis(DPA)] moieties using carbodiimide coupling chemistry. The complexation of zinc (Zn2+) ion is achieved by exposing the modified pore to a solution of zinc chloride to form bis(Zn2+¿DPA) complexes. Micellar Liquid Chromatography:  A Worthy Technique for the Determination of β-Antagonists in Urine Samples

Several beta-antagonists (acebutolol, atenolol, celiprolol, labetalol, metoprolol, nadolol, propranolol) were determined in urine samples with fluorometric detection after direct injection, in less than 15 min, with a micellar mobile phase of 0.1 M sodium dodecyl sulfate (SDS), 15% propanol, and 1% triethylamine at pH 3. The limits of detection (38 criterion) were usually between 3 and 30 ng/mL. The addition of propanol and triethylamine and the reduction of the pH of the mobile phase improved the efficiency of the chromatographic peaks that was rather low in pure micellar eluents. Performance of amines as silanol suppressors in reversed-phase liquid chromatography

In reversed-phase liquid chromatography, cationic basic compounds yield broad and asymmetrical peaks, as a result of their ionic interaction with the anionic free silanol groups present in the silica-based stationary phases (commonly derivatised with C18 groups). A simple way to improve the peak shape is the addition to the hydro-organic mobile phase of a reagent (usually called additive) with cationic character. 1. The effect of histamine and histamine H1- and H2-receptor agonists on isolated myometrium strips of premenopausal women has been examined. The effect of acetylcholine was also determined. 2. Histamine, 2-pyridylethylamine, 4-methylhistamine and acetylcholine, but not dimaprit, produced a concentration-related contractile response in human isolated myometrial strips. The aim of the study was to investigate the influence of 40 mg of the beta-blocker penbutolol (Betapressin TM; Hoechst Ltd., Frankfurt/Main) in comparison to placebo on the insulin consumption on the blood sugar profile in twelve insulin-dependent diabetes (IDDM) patients. The patients were treated with penbutolol and placebo for a period of three days, and then were examined with the help of the glucose-controlled insulin infusion system. The blood sugar profile and insulin consumption over a 24 hour period was not affected by either penbutolol or placebo, nor could any changes be measured in these parameters when measured after food intake. Plerixafor with and without chemotherapy in poor mobilizers: results from the German compassionate use program.

The CXCR4-inhibitor plerixafor mobilizes hematopoietic stem cells amplifying the effects of granulocyte-CSF (G-CSF). Before approval plerixafor was used in a compassionate use program (CUP) for patients who failed a previous mobilization. In the German CUP 60 patients from 23 centers (median age 56.5 years (2-75)) were given 240 μg/kg plerixafor SC 9-11 h before apheresis. A total of 78.3% (47/60) received G-CSF for 4 days before plerixafor administration; 76.6% of those (36/47) yielded at least 2.0 × 10(6) CD34(+) cells/μL.
