A Long Journey into the Investigation of the Structure–Dynamics–Function Paradigm in Proteins through the Activities of the Palermo Biophysics Group
2022
An overview of the biophysics activity at the Department of Physics and Chemistry Emilio Segrè of the University of Palermo is given. For forty years, the focus of the research has been on the protein structure–dynamics–function paradigm, with the aim of understanding the molecular basis of the relevant mechanisms and the key role of solvent. At least three research lines are identified; the main results obtained in collaboration with other groups in Italy and abroad are presented. This review is dedicated to the memory of Professors Massimo Ugo Palma, Maria Beatrice Palma Vittorelli, and Lorenzo Cordone, which were the founders of the Palermo School of Biophysics. We all have been, directl…
Genetic manipulation of spiramycin-producing strain Streptomyces ambofaciens ATCC 23877 by φC31 Att/Int system-based vectors results in a significant…
The genus Streptomyces produces about two-thirds of naturally occurring antibiotics and many other biologically active secondary metabolites. The genetic manipulation of Streptomycetes is often labor and time intensive due to their large genome and complex development. Much progress has been made to develop gene transfer methods useful to construct antibiotic-producing strains with improved properties. The φC31 Att/Int system is an integration system that has been widely used to produce stable recombinants in Actinomycetes and its integration site is an attB site located in a pirin-like gene (pirA) of the bacterial chromosome. In this study we demonstrate that the integration of φC31 DNA in…
Bisexual desires for more than one gender as a challenge to normative relationship ideals
2018
Normative western understandings of intimate relationships continue to draw upon the discourses of romantic love and the ideal of finding ‘the one’ who meets all our romantic and sexual needs. As desire is not sexually or emotionally exclusive, even people in normative relationships have to make sense of desires beyond the monogamous ideal. Bisexual people engage in these negotiations from a challenging cultural position. As a desire for more than one gender, bisexuality is persistently culturally associated with wavering desire, promiscuity and multiple partners. In light of these cultural conditions, I explore how Finnish bisexual women – and their (ex-)partners of various genders who do …
Similar and equal relationships? Negotiating bisexuality in an enduring relationship
2015
In the public debate in Finland, same-sex couples’ right to legal recognition is routinely defended by stressing their sameness to heterosexual couples within the discourse of romantic love. This article explores how bisexual women and their partners use these discourses. The five couple interviews were analyzed by implementing discourse analysis. The results highlight how, when taking positions within the discourse of the enduring couple relationship, the interviewees drew on the discourse of romantic love. Woman’s bisexuality disappeared easily in this talk. Although it seemed effortless at first sight, negotiations and affective tensions arose when the interviewees tried to fit their re…
Itsemurhan taide : itsemurhan roolit modernissa ja jälkimodernissa taiteessa
2011
Tutkielma kartoittaa itsemurhan esiintyvyyttä modernissa ja jälkimodernissa taiteessa ja pyrkii luomaan typologian niistä erinäisistä rooleista, joissa itsemurhan representaatio tällä reilun sadan vuoden aikajanalla esiintyy. Itsemurhan näkyvyydestä taiteessa tehty aiempi tutkimus on vähäistä ja painottuu modernismia edeltäneeseen taiteeseen, joten tutkielma pyrkii muodostamaan kokonaiskuvaa niukasti tutkitusta aihepiiristä.
MTOR inhibitor-based combination therapies for pancreatic cancer
2018
Background: Although the mechanistic target of rapamycin (MTOR) kinase, included in the mTORC1 and mTORC2 signalling hubs, has been demonstrated to be active in a significant fraction of patients with pancreatic ductal adenocarcinoma (PDAC), the value of the kinase as a therapeutic target needs further clarification. Methods: We used Mtor floxed mice to analyse the function of the kinase in context of the pancreas at the genetic level. Using a dual-recombinase system, which is based on the flippase-FRT (Flp-FRT) and Cre-loxP recombination technologies, we generated a novel cellular model, allowing the genetic analysis of MTOR functions in tumour maintenance. Cross-species validation and pha…
Double copies of blaKPC-3::Tn4401a on an IncX3 plasmid in Klebsiella pneumoniae successful clone ST512 from Italy
2015
ABSTRACT A carbapenem-resistant sequence type 512 (ST512) Klebsiella pneumoniae carbapenemase 3 (KPC-3)-producing K. pneumoniae strain showing a novel variant plasmid content was isolated in Palermo, Italy, in 2014. ST512 is a worldwide successful clone associated with the spread of bla KPC genes located on the IncFIIk pKpQIL plasmid. In our ST512 strain, the bla KPC-3 gene was unusually located on an IncX3 plasmid, whose complete sequence was determined. Two copies of bla KPC-3 ::Tn 4401a caused by intramolecular transposition events were detected in the plasmid.
Effect of RNS60 in amyotrophic lateral sclerosis: a phase II multicentre, randomized, double-blind, placebo-controlled trial
2022
Background and purpose: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with limited treatment options. RNS60 is an immunomodulatory and neuroprotective investigational product that has shown efficacy in animal models of ALS and other neurodegenerative diseases. Its administration has been safe and well tolerated in ALS subjects in previous early phase trials. Methods: This was a phase II, multicentre, randomized, double-blind, placebo-controlled, parallel-group trial. Participants diagnosed with definite, probable or probable laboratory-supported ALS were assigned to receive RNS60 or placebo administered for 24 weeks intravenously (375 ml) once a week and via nebul…
