Abstract LiChrossorb Diol® packing has been to be well-suited for the separation of proteins and enzymes according to a size-exclusion mechanism in a mean molecular-weight (MW) range between 10,000 and 100,000. Loadability of a small bore column of 6 mm I.D. (A) and a large-bore column of 23.5 mm I.D. (B), both of 250 mm in length, were examined. Several chloroformic extracts containing some sesquiterpene lactones from autochtonous plants of Parthenium hysterophorus, collected in the province of Salta (Argentina), were separated and identified by high performance reversed-phase liquid chromatography with C18 column and propanol-water mobile phases, using gradient elution and concentrations of propanol larger than 10% (v/v). Abstract The reversed-phase chromatography of proteins by gradient elution with acidic, low-ionic-strength aqueous—organic eluents is often associated with losses of the biological activity of the protein. In this study, the enzymatic activities of catalase, horseradish peroxidase and pepsin were examined under static and dynamic column conditions on non-porous, monodisperse 1.5-μm reversed-phase silicas with various n -alkyl ligands. Flow-injection spectrophotometric determination of amino acids based on an immobilised copper(II) -zincon system

Abstract The flow-injection spectrophotometric determination of different amino acids was carried out by reaction with copper(II) ions entrapped in a polymeric material and filling a packed-bed reactor; the released copper(II), complexed with the amino acid, reacted with zincon in a basic medium producing a blue colour that was monitored at 600 nm. The method was applied to determine the contents of different amino acids in pharmaceutical formulations. The calibration graph for glycine was linear over the range 0.5–20 μg ml −1 with a relative standard deviation of 0.8% ( n = 6) at 10 μg ml −1 and a sample throughput of 108 h −1 .

γ-Oryzanol and tocopherol contents in residues of rice bran oil refining

Rice bran oil (RBO) contains significant amounts of the natural antioxidants γ-oryzanol and tocopherols, which are lost to a large degree during oil refining. This results in a number of industrial residues with high contents of these phytochemicals. With the aim of supporting the development of profitable industrial procedures for γ-oryzanol and tocopherol recovery, the contents of these phytochemicals in all the residues produced during RBO refining were evaluated. The samples included residues from the degumming, soap precipitation, bleaching earth filtering, dewaxing and deodorisation distillation steps. Inhibition of cyclodextrins on α-galactosidase.

This work successfully investigated the effects of different influential factors and hydrophobic cavities of cyclodextrins (CDs) on α-galactosidase (α-Gal) by detecting α-Gal activity. The highest inhibitory concentration of three kinds of CDs (α-, β-, and γ-CD) on α-Gal was 10mM. Moreover, the highest inhibition of α-Gal was obtained under the following conditions: reaction time of 90min, temperature of 30°C, and pH 6.0. Compared with other CDs, β-CD showed more ability to interact with α-Gal due to its appropriate cavity geometric dimensions. The phenomenon of co-nonsolvency, i.e. the formation of nonsolvents by mixing two solvents, was studied for the system N,N-dimethylformamide/cyclohexane/polystyrene (DMF/CH/PS) which is characterized by the fact that the components of the mixed solvent are only incompletely miscible below 48,6°C. A number of quasibinary sections (stepwise addition of the second solvent to given solutions of the polymer in the first solvent) for a sample (M = 110000) of high molecular uniformity yielded a cloud point surface which gives “island curves” (closed solubility gaps) for constant temperatures between the critical temperature of DMF/CH and the top of the cloud point surface (51,3°C). Optical tweezing electrophoresis of isolated, highly charged colloidal spheres

Abstract We report on a novel optical tweezing experiment measuring the electrophoretic mobility μ of highly charged spherical particles suspended in water at volume fractions Φ≤2×10 −7 . At deionised conditions μ =2.5×10 −8 m 2 V −1 s −1 , it decreases further upon increasing the salt concentration c . We compare our data to measurements at larger Φ, where generally much larger μ and a qualitatively different dependence on the salt concentration are observed. To characterise the dependence on Φ we performed measurements at no added salt.
