CK2β-regulated signaling controls B cell differentiation and function

Serine-Threonine kinase CK2 supports malignant B-lymphocyte growth but its role in B-cell development and activation is largely unknown. Here, we describe the first B-cell specific knockout (KO) mouse model of the β regulatory subunit of CK2. CK2βKO mice present an increase in marginal zone (MZ) and a reduction in follicular B cells, suggesting a role for CK2 in the regulation of the B cell receptor (BCR) and NOTCH2 signaling pathways. Biochemical analyses demonstrate an increased activation of the NOTCH2 pathway in CK2βKO animals, which sustains MZ B-cell development. Cattle babesiosis is a tick-borne disease transmitted by haemoparasites belonging to the phylum of Apicomplexa, such as Babesia bovis and B.bigemina. The pathology affects cattle mainly in tropical and subtropical areas, but also in Europe, strongly reducing meat and milk production. The study performed during these three years of PhD was addressed to the molecular characterization of Italian B.bigemina strains isolated from infected animals, with attention to the genes codifying for surface antigens, putative candidates for vaccine and diagnostic tools development. Gradual ocean warming and marine heatwaves represent major threats for marine organisms already facing other anthropogenic-derived hazards, such as chemical contamination in coastal areas. Abstract The persistent pollutants polybrominated diphenyl ethers (PBDEs) have been demonstrated to produce several negative effects on marine organisms. Although Mytilus galloprovincialis was extensively studied as model system, the effects of PBDEs on the innate immune system of mussels remains unclear. In this study, except for the control treatment, specimens of M. galloprovincialis were fed with microalgae treated with increasing concentrations of PBDEs (maximum level 100 ng L−1 of BDE-47 per day).
