H2O2 resistant mesoangioblast clone isolation with a distinct survival advantage in vitro and in vivo

2017

The release of molecules from damaged tissues stimulates both resident and circulating stem cells to initiate a tissue repair programme. 1 However, during transplantation procedures the therapeutic efficacy of stem cells is compromised by reduced homing capability towards the target site.2,3 Furthermore, cell survival is very low and many studies focused on improving cell viability upon implantation. In this study, we performed in vitrosevere oxidative stress to select some more resistant mouse mesoangioblasts, vessel-associated progenitor stem cells endowed with the ability of multipotent mesoderm differentiation. We found that the selected subpopulation retains selfrenewal and myogenic di…

Sperm p53 concentration: a potential new biomarker for environmental pollution. Preliminary data.(EcoFoodFertility Project)

2019

Study question:We evaluated the p53 protein concentration in spermcells to test whether environmental pollution can affect expression levels of this protein. Summary answer:We found a significant difference in p53 levels between two homogeneous groups by age and lifestyle, residents in two areas with different environmental impact. What is known already:TheWorld HealthOrganization (WHO) has placed among its priority objectives the understanding of the relationships between the sources of pollution and the effects on human health, which they represent a cause of surprisingly high mortality and morbidity. International scientific literature shows that strong environmental impact may jeopardiz…

Pippin protein expression changes during cell differentiation

2008

PIPPin is a CSD-containing protein with the ability to interact both with mRNAs encoding histone variants and chromatin. A major fraction of chromatin-bound PIPPin is sumoylated and sumoylation seems to be controlled by thyroid hormones, both in vivo and in vitro. We studied its expression in different tissues and cell lines and even in tumor cells and found that, even if more expressed in the brain respect to other tissues of the adult rat, it is also expressed in brain tumors and in cell lines as different as kidney NRK cells and PC12. The expression of the protein is strongly increased by treatments that induce differentiation, such as treatment of PC12 with NGF. We also found an increas…

Porous PLLA scaffolds are optimal substrates for internal colonization by A6 mesoangioblasts and immunocytochemical analyses

2009

In the present paper, mouse mesoangioblasts were seeded onto bidimensional matrices and within three-dimensional porous scaffolds of poly(L-lactic acid) (PLLA), in the presence or absence of type I collagen coating, observed under the scanning electron microscope, and tested for their adhesion, survival and proliferation. Immunolocalization of Hsp70, an abundant and ubiquitous intracellular protein in these cells, was also performed in sectioned cell-containing scaffolds under the confocal fluorescence microscope to check whether "in situ" analysis of intracellular constituents was feasible. The data obtained show that PLLA films allow direct cell adhesion and represent an optimal support f…

Apoptosis rate of cumulus cells can be considered as an indicator for the selection of embryos to improve ongoing pregnancy and implantation rates.

2014

Cumulus cells apoptosis rate an adjunct to morphology evaluation for the embryo selection on day 3 could be considered a new tool, compared with embryo selection by morphology alone, to select the embryos with higher implantation potential to increase the clinical outcomes after ICSI. Several studies have demonstrated a lower cumulus cell apoptotic rates in women who achieved pregnancy compared with women who did not become pregnant after ICSI. A prospective randomized observational study on 76 ICSI patients was performed before Ovum Pick-Up. Patients were randomized into either the control group (embryo selection by morphology only, A group: 48 patients) or the treatment group (morphology …
