Versuche zur somatischen Beeinflussung der Organbildung pflanzlicher Embryonen
1955
Experiments on influencing cotyledon formation in embryos ofEranthis hiemalis are reported. By treating the seeds with 100–2000 ppm solutions of 2,4-dichlorophenoxyacetic acid, 2,4,5-trichlorophenoxyacetic acid, 2-methyl-4-chlorophenoxyacetic acid, α-naphthylacetic acid and even isopropyl N-phenyl carbamate one can obtain significant increases in twinning and in cotyledon anomalies (syncotyly and pleiocotyly of all degrees). Embryos can be successfully treated until approximately 2 weeks after initial opening of the follicles. X-radiations achieve the same effect, with doses of 2000–4000r corresponding to the treatments with 1000–2000 ppm 2,4-D-solutions. By using weaker concentrations and …
Funktion der Spermien in inkompatiblen Kreuzungen der StechmückeCulex pipiens L.
1972
After irradiating males of the incompatible crossC. pipiens ‘Hamburg’ ♀♀×C. pipiens ‘Paris’ ♂♂ with X-ray doses between 1 kR and 40 kR, it could be shown that in this cross the sperm has only the function of activating the egg. All the hybrid embryos develop from the haploid female pronucleus.
MicroRNA-30d deficiency during preconception affects endometrial receptivity by decreasing implantation rates and impairing fetal growth.
2019
Background Maternal–embryonic crosstalk between the endometrium and the preimplantation embryo is required for normal pregnancy. Our previous results demonstrated that maternal microRNAs secreted into the endometrial fluid, specifically miR-30d, act as a transcriptomic regulator of the preimplantation embryo by the maternal intrauterine environment. Objective To investigate the reproductive and fetal effects of murine miR-30d deficiency at the maternal–embryonic interface according to the origin of its maternal or embryonic default. Study Design A miR-30d knockout murine model was used as the animal model to investigate the impact of maternal and/or embryonic origin of miR-30d deficiency on…
Optimal Timing for Oocyte Denudation and Intracytoplasmic Sperm Injection
2011
Objectives. To analyze the impact of oocyte denudation and microinjection timings on intracytoplasmic sperm injection (ICSI) outcomes.Study Design. We included ICSI cycles with the following parameters: rank 1 or 2, female age years, male factor infertility, long protocol using GnRH agonist and rFSH for ovarian stimulation, and use of freshly ejaculated sperm (). Several ICSI parameters were analyzed according to the time between oocyte retrieval and denudation () and the time between denudation and ICSI () using a statistical logistic regression analysis.Results. Neither nor had a significant influence on the Metaphase II (MII) rate but the fertilisation rate (FR) showed a significant impr…
Gene polymorphisms and HLA-G expression in spontaneous abortions
2015
Abstract Introduction HLA-G and HLA-E are claimed to play a role in establishing maternal–fetal immune tolerance and in maintaining pregnancy. The presence of polymorphism in the HLA-G gene could cause a deficient or excessive expression of the HLA-G and HLA-E molecules. These anomalies could eventually cause pregnancy losses. Materials and methods Clinical study. A total of 90 patients were included in this study. These patients suffered spontaneous abortions between weeks 6 and 11 of pregnancy. We have analysed the most important polymorphisms of the HLA-G gene through different genetic studies and HLA-G and HLA-E expression through immunostaining in human cytotrophoblast cells from first…
Embryo classification method based in time-lapse analysis is associated with blastocyst quality
2012
groups: 87.7%(10434/11893), 87.4%(2674/3060), 80.2%(1350/1683) and 81.0%(634/783). Abnormal fertilization rate was significantly lower in 9598% group(6.9%,54/783) than other groups(16.1%,1918/11893; 13.8%,421/ 3060; 11.4%,192/1683). Good embryo, pregnancy and implantation rates were no significant differences among four groups(48.9%,61.6%,43.5%; 48.9%,62.1%,43.5%; 50.0%,57.6%,42.3%; 53.1%,62.5%,45.7%). CONCLUSION: Sperm morphology does not affect normal fertilization, embryonic development and clinical outcome.
P–231 Transnational oocyte donation program between Italy and Spain based on transport of vitrified oocytes: a five-years experience
2021
Abstract Study question What is the clinical efficacy of an oocyte donation program based on the transportation of vitrified oocytes between two countries? Summary answer The transnational oocyte donation program is efficient, safe and comparable to other strategies (transport of frozen sperm and embryos). What is known already Egg donation represents a valid treatment strategy for women who have exhausted their ovarian function and it has considerably increased in the last years. In Italy, egg donation is allowed after the judgment of the Constitutional Court n. 162 in 2014 but no reimbursement for the donors is provided. For this reason, the number of voluntary donors is irrelevant. There…
Postnatal exposure to N-ethyl-N-nitrosurea disrupts the subventricular zone in adult rodents
2010
N-ethyl-N-nitrosurea (ENU), a type of N-nitrous compound (NOC), has been used as inductor for brain tumours due to its mutagenic effect on the rodent embryo. ENU also affected adult neurogenesis when administered during pregnancy. However, no studies have investigated the effect of ENU when exposured during adulthood. For this purpose, three experimental groups of adult mice were injected with ENU at different doses and killed shortly after exposure. When administered in adult mice, ENU did not form brain tumours but led to a disruption of the subventricular zone (SVZ), an adult neurogenic region. Analyses of the samples revealed a reduction in the numbers of neural progenitors compared wit…
Swelling and death of neuronal cells by lactic acid.
1993
Lactacidosis occurring in cerebral ischemia or trauma is a major mechanism of cytotoxic brain edema and brain damage. Respective effects of lactacidosis were currently analyzed in vitro by employment of the murine neuronal cell line, Neuro-2A, in order to obtain a better understanding of specific mechanisms underlying cell swelling and cell death in comparison with glial cells. The cells were suspended in a physiological medium in the presence of lactic acid at increasing concentrations. Levels of acidosis reaching from pH 6.8-5.6 were obtained while other parameters, such as osmolarity and electrolyte concentrations, were maintained in the physiological range. Assessment of cell swelling a…
Dexamethasone treatment of naïve organ of Corti explants alters the expression pattern of apoptosis-related genes.
2009
Dexamethasone treatment of organ of Corti explants challenged with an ototoxic level of an inflammatory cytokine modulates NFkappaB signaling and the expression levels of both pro-and anti-apoptosis-related genes. It is not known if naïve organ of Corti explants will respond in a similar manner to treatment with a corticosteroid. This study examines the response of naïve organ of Corti explants to treatment with dexamethasone.Three-day-old rat organ of Corti explants were cultured for 1, 2, or 4 days. Four-day in vitro cultures were fixed, stained with FITC-phalloidin and hair cells were counted. ELISA was performed on 2-day cultures to determine the levels of phosphorylated nuclear factor …
