Mesenchymal Stem Cells Improve Motor Functions and Decrease Neurodegeneration in Ataxic Mice

The main objective of this work is to demonstrate the feasibility of using bone marrow-derived stem cells in treating a neurodegenerative disorder such as Friedreich's ataxia. In this disease, the dorsal root ganglia of the spinal cord are the first to degenerate. Two groups of mice were injected intrathecally with mesenchymal stem cells isolated from either wild-type or Fxntm1Mkn/Tg(FXN)YG8Pook (YG8) mice. As a result, both groups presented improved motor skills compared to nontreated mice. High levels of neuropeptide Y (NPY) are found in basal ganglia where it is co-localised with somatostatin (SOM) and nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH/d) in a population of striatal GABA containing interneurones. Although alterations occur in the levels of various neuropeptides in basal ganglia in Parkinson’s disease (PD), it is not known whether NPY is affected. Inhibition of nitric oxide synthase influences the activity of striatal neurons in the rat

The activity of single units in the striatum of urethane-anesthetized rats was recorded before and after the systemic administration of 7-nitro-indazole (7-NI; 50 mg/kg intraperitoneally), a selective inhibitor of neuronal nitric oxide (NO) synthase. Two neuronal types were clearly distinguishable electrophysiologically, on the basis of either discharge frequency pattern or features of the individual spike waveform (spike duration, negative phase/total duration ratio, and negative phase/total amplitude ratio). Basal ganglia calcifications and ALS syndrome.

We report the case of a patient with idiopathic hypoparathyroidism and unusually large symmetrical calcifications in the basal ganglia, thalami, cerebellar hemispheres and brainstem, who clinically presented an ALS-like syndrome. We discuss the possible role of abnormal calcium metabolism in the pathogenesis of motoneuron disease.

Gastric acid secretory responses induced by peptone are mediated by capsaicin-sensitive sensory afferent neurons

The involvement of capsaicin-sensitive afferent neurons in modulating acid-secretory responses to peptone, a product of protein digestion, has been investigated in the continuously perfused stomach of the urethan-anesthetized rat. Systemic neonatal pretreatment with capsaicin, which destroys primary afferent neurons, does not modify basal levels of acid secretion. Acid responses to intragastric perfusion with isotonic (0.5, 1, and 2.4%) or hypertonic (10 and 20%) solutions of peptone were reduced in capsaicin-treated rats. The role of choline in the release of acetylcholine in isolated hearts.

1. The concentrations of acetylcholine, choline and noradrenaline were estimated in the perfusate (overflow) of isolated hearts of chickens, cats, rabbits and guinea pigs. Neurotransmitter release was evoked by stimulation of both vagus nerves and by direct stimulation of the heart (field stimulation). 2. In the absence of exogenous choline and physostigmine, field stimulation at 20 Hz for 20 min caused an overflow of acetylcholine from the hearts of the 4 species investigated. Distribution of dopamine-beta-hydroxylase—like immunoreactivity in the rat pineal organ

The aim of the present investigation was to study the distribution in the rat pineal gland of dopamine-beta-hydroxylase (DBH) which is essential for the formation of the melatonin synthesis-regulating substance noradrenaline (NA). Electrophysiological characterization of the pineal gland of golden hamsters.

In one of the most-widely used species in pineal gland research, the strongly photoperiodic golden hamster, Mesocricetus auratus, no electrophysiological data on pinealocytes are currently available. To fill this gap, in the present study 185 spontaneously active pinealocytes from male golden hamsters were recorded extracellularly, both during day- and night-time (light: dark cycle 12 ∶ 12, lights on at 07.00 h). As in other species, pinealocytes exhibited action potentials of 1–2 ms duration. Principal neurons projecting to the pineal gland in close association with small intensely fluorescent cells in the superior cervical ganglion of rats

The localization in the superior cervical ganglia (SCG) of small, intensely fluorescent (SIF) cells and of principal nerve (PN) cells innervating the pineal gland was examined in adult male Sprague-Dawley rats. PN cells were demonstrated by means of the retrograde neuron-tracing method using the fluorescent tracer Fluoro-Gold (FG) injected into the pineal gland.
