Abstract The improvement of nucleic acid probes as vital molecular engineering devices will cause a noteworthy contribution to developments in bioimaging, biosensing, and disorders diagnosis. The molecular beacon (MB) which was designed by Tyagi and Kramer in 1996, are loop-stem hairpin-designed oligonucleotides armed with a quencher and a dye (also named reporter groups) at the 3′ or 5′ ends. This construction allows that MBs in the absence of their target complementary molecules do not fluoresce. The aquatic larvae of non-biting midges (Chironomidae, Diptera) contain a variety of Hb proteins in their hemolymph that enable them to survive in an anoxic environment (1). In Chironomus thummi thummi, 12 different Hb variants have been identified and their amino acid sequences determined (2). Based on these primary structures, the evolutionary relationships between the five monomeric and the s e v e n dimeric Hb proteins have been deduced (2). The two groups are thought to have evolved in two different lineages which separated more than 255 million years ago.
Oxygen-induced changes in hemoglobin expression in Drosophila

The hemoglobin gene 1 (dmeglob1) of the fruit fly Drosophila melanogaster is expressed in the tracheal system and fat body, and has been implicated in hypoxia resistance. Here we investigate the expression levels of dmeglob1 and lactate dehydrogenase (a positive control) in embryos, third instar larvae and adult flies under various regimes of hypoxia and hyperoxia. As expected, mRNA levels of lactate dehydrogenase increased under hypoxia. We show that expression levels of dmeglob1 are decreased under both short- and long-term hypoxia, compared with the normoxic (21% O2) control. Carotenoid-based integumental coloration is often associated with individual performance in various animals. This is because the limited amount of the pigment has to be allocated to different vital functions. However, most of the evidence for the carotenoid-based trade-off comes from vertebrate studies, and it is unclear if this principle can be applied to insects. Tenascin is a large oligomeric glycoprotein of the extracellular matrix. Its location is limited in innervated muscle tissues. We investigated immunohistologically, using two monoclonal antibodies (mab) against Tenascin, biopsied denervated human muscle of children and adults. Tenascin was present in the interstitial space among denervated muscle fibres. Accumulation of Tenascin in denervated adult muscle tissue was frequent, accumulation in denervated muscle tissue of children was sparse and weak. The two antibodies reacted correspondingly. Equilibrium (pH-metric) and spectroscopic (1H,13C, and 119Sn NMR and 119Sn Mossbauer) studies were performed to characterize the interaction of the dimethyltin(IV) cation with glycine, glycyl-glycine (Gly-Gly), imidazole-4-acetic acid, histamine, histidine, glycyl-histamine, glycyl-histidine (Gly-His), and β-alanyl-histidine (carnosine). For histamine and glycyl-histamine (having only nitrogen donor atoms) no complex formation was detected. A simple, large-scale synthesis of a cytokine, methionine-enkephalin, Tyr-Gly-Gly-Phe-Met, has been elaborated. Classical solution peptide chemistry methods without protection of amino acid side-chain functions and 1+(2+2) segment condensation were used. A nine-step synthesis from commercial amino acid derivatives was developed with yields ranging from 86% to 99%, averaging 92%. The purity of all intermediates was found to be 99.0-100% by HPLC. The process has been used to prepare greater than 150 g quantities of the pentapeptide as a monohydrate of 100% purity. Hydantoin formation was observed during saponification of Boc-Tyr-Gly-Gly-Phe-Met-OMe and minimised.

Abstract Glycosyl-N-allyl carbamates, obtained by reaction of anomerically unprotected saccharides with allyl isocyanate, can be activated by an electrophile-induced cyclisation and reacted with glycosyl acceptors to form the corresponding oligosaccharides By this method the mucin core 2 trisaccharide2 has successfully been synthesized. Due to the mild glycosylation conditions even 1-O-acetyl protected glycosyl acceptors can be used. This was demonstrated in the synthesis of a 1,6-linked glucosyl trisaccharide whereby a reptitious glycosylation strategy could be applied. 1. Dedicated to the memory of Professor Akira Hasegawa.
