Clean method for the simultaneous determination of propyphenazone and caffeine in pharmaceuticals by flow injection Fourier transform infrared spectr…
1997
A procedure is proposed for the simultaneous FTIR determination of propyphenazone (PFZ) and caffeine (CAF) in pharmaceuticals. The method involves the dissolution of the active principles in CHCl 3 , followed by filtration of sample solutions to remove the excipients. PFZ is then determined by absorbance measurements at 1595 cm - 1 , using a baseline established between 2000 and 890 cm - 1 , and CAF by using the first-derivative values at 1712 cm - 1 , using solutions of PFZ and CAF for external calibration. The method was applied in both the stopped-flow and flow-injection modes, providing precise and accurate results for the analysis of real samples. The incorporation of a distillation un…
Pixel-to-pixel correlation between images of absolute ATP concentrations and blood flow in tumours
1992
Iodo(14C-)antipyrine autoradiography and imaging bioluminescence have been combined to obtain pixel-to-pixel correlations between absolute values for local blood flow and ATP concentrations at a microscopical level within designated areas in hamster melanomas. Positive pixel-to-pixel correlations were obtained in 4 of 6 tumours. Both flow and ATP values were less in mostly necrotic than in mostly viable tumour regions. The data provide evidence for the energetic state of cancer cells being strongly influenced by the efficiency of tumour microcirculation in several but not in all malignancies investigated. Lysosomal degradation of the carboxydextran shell of coated superparamagnetic iron oxide nanoparticles and the fate of professional phagocytes
2010
Contrast agents based on dextran-coated superparamagnetic iron oxide nanoparticles (SPIO) are internalized by professional phagocytes such as hepatic Kupffer cells, yet their role in phagocyte biology remains largely unknown. Here we investigated the effects of the SPIO ferucarbotran on murine Kupffer cells and human macrophages. Intravenous injection of ferucarbotran into mice led to rapid accumulation of the particles in phagocytes and to long-lasting increased iron deposition in liver and kidneys. Macrophages incorporate ferucarbotran in lysosomal vesicles containing α-glucosidase, which is capable of degrading the carboxydextran shell of the ferucarbotran particles. Intravenous injectio…
Evidence of a specialized transport mechanism for the intestinal absorption of baclofen
1989
Absorption of the spasmolytic drug baclofen in three selected intestinal segments of living anaesthetized rats in situ, is shown to be a specialized transport mechanism obeying Michaelis-Menten kinetics. Equation parameters were calculated through different procedures, whose features are discussed. A computer method based on the integrated form of Michaelis-Menten equation which reproduces the entire time course of drug absorption from the data found in three intestinal perfusion series at different initial concentrations, yielded Vm and Km values of 12.0 mg h-1 and 8.0 mg, respectively, in the mean segment of the small intestine, a rather selective absorption site for baclofen. Lesser but …
