Effects of pH and the presence of micelles on the resolution of diuretics by reversed-phase liquid chromatography

2004

A comparative study on the performance of two RPLC modes on the separation of 18 diuretics with diverse acid-base behaviour (acetazolamide, althiazide, amiloride, bendroflumethiazide, benzthiazide, bumetanide, canrenoic acid, chlorothiazide, chlorthalidone, ethacrynic acid, furosemide, hydrochlorothiazide, piretanide, probenecid, spironolactone, triamterene, trichloromethiazide and xipamide) was carried out. A conventional octadecylsilane column and acidic acetonitrile-water mobile phases, in the absence and presence of micelles of the anionic surfactant sodium dodecyl sulphate (SDS), were used. The effects of pH and the modifiers acetonitrile and SDS on peak asymmetry, efficiency, selectiv…

Chromatographic monitoring of diuretics in urine samples using a sodium dodecyl sulphate—propanol micellar eluent

1994

Abstract The effect of a varying pH, in the range 3−7, on the retention of several diuretics eluted with a sodium dodecyl sulphate (SDS) micellar mobile phase and with UV detection was studied. Significant changes in the capacity factors ( K′ of the diuretics bumetanide, ethacrynic acid, furosemide, probenecid and xipamide were observed. The protonation constants of these compounds were calculated from the k′ data. A study was also performed to select the optimum composition of the mobile phase (pH, concentration of SDS and nature and concentration of alcohol) for the separation of the above diuretics and the diuretics amiloride, bendroflumethiazide, chlorthalidone, hydrochlorothiazide, spi…

Chromatographic determination of diuretics in urine samples using hybrid micellar mobile phases with fluorimetric detection

1998

Abstract A rapid and simple chromatographic procedure, that uses hybrid micellar mobile phases of sodium dodecyl sulphate (SDS), propanol and phosphate buffer at pH 3, and fluorimetric detection ( λ exc =270 nm, λ em =430 nm), is reported for the determination of mixtures of seven diuretics of diverse efficacy (amiloride, bendroflumethiazide, bumetanide, furosemide, hydroflumethiazide, piretanide and triamterene), in urine samples. The application of a sequential optimization procedure for the resolution of the mixtures of the diuretics was problematic. However, their separation was achieved through the use of an interpretive optimization procedure based on the accurate description of the r…

A hybrid bioinspired material with manganese(II) complexes grafted on the surface of a mesostructured porous silica is investigated. The Mn sites mimic the manganese-dependent dioxygenase (MndD), which is an enzyme that catalyses the oxidation of catechol derivatives. The metal complexes were introduced in the silica using a dinuclear complex [Mn2L2(Cl)2(μ-Cl)2] as a precursor with a clickable ligand N,N′-bis[(pyridin-2-yl)methyl]prop-2-yn-1-amine (L). Azide moieties covalently grafted on MCM-41 type mesoporous silica were utilised to anchor the manganese complex through Huisgen cycloaddition using CuBr(PPh3)3 as a catalyst. A second functional group – trimethylsilyl or pyridine—was grafted…

New nitrene functionalizations onto sidewalls of carbon nanotubes and their spectroscopic analysis

2010

Abstract The reactivity of p -toluenesulfonyl, methylsulfonyl and trimethylsilyl nitrene, derived from the corresponding azides, was studied towards single-walled carbon nanotubes (SWCNT) prepared by electric arc or HiPCO (High-pressure CO conversion) methods. The functionalized SWCNTs were analyzed by Raman, IR, and VIS/NIR spectroscopy. The spectroscopic results indicated that covalent modification of the SWCNTs was successful. While the IR measurements gave evidence of successful reaction in all studied cases, the Raman measurements indicated differences in the reactivity of the two tube types and between different nitrenes. VIS/NIR spectrum was measured for reaction with p -toluenesulfo…

Combined effect of solvent content, temperature and pH on the chromatographic behaviour of ionisable compounds.

2008

The organic solvent content and the pH in the mobile phase are the usual main factors in reversed-phase liquid chromatographic separations, owing to their strong effects on retention and/or selectivity. Temperature is often neglected. However, even in cases where the impact of this factor on selectivity is minor, the reduction in analysis time is still an interesting reason to consider it. In addition, ionisable compounds may exhibit selectivity changes, owing to the interaction of organic solvent and/or temperature with pH. The separation of ionisable compounds (nine diuretics: bendroflumethiazide, benzthiazide, bumetanide, chlorthalidone, furosemide, piretanide, probenecid, trichloromethi…

Synthesis of new asymmetric xanthene dyes via catalyst-free SNAr with sulfur nucleophiles

2014

Addition of a single functional handle to the tricyclic moiety of fluorescein results in asymmetric xanthene dyes. Our synthesis of a new class of asymmetric xanthenes proceeds via an unusual SNAr with sulfur nucleophiles on electron rich aromatic xanthenes scaffolds in the absence of a metal catalyst. The resulting 3'-thioethers exhibit high photostability and are conveniently converted into reactive dyes for macromolecule labelling.

π-Electron delocalization in 2-benzoyl-5-phenylpyrazolidin-3-one

2018

Abstract The crystal and molecular structures of 2-benzoyl-5-phenylpyrazolidin-3-one have been characterized by X-ray diffraction along with density functional theory studies. Cinnamic acid chloride was reacted with benzhydrazide, yielding 2-benzoyl-5-phenylpyrazolidin-3-one. This product was formed in the transformation comprising the nucleophilic addition of benzhydrazide to the styryl fragment of the α,β-unsaturated arrangement and subsequent cyclization. The molecule contains two benzene rings and one five-membered heterocyclic ring with an N–N single bond. The five-membered ring is composed of three atoms of sp 3 hybridization and two atoms of sp 2 hybridization, which cause the flatte…

The effect of amphiphilic compounds on the secretion of levansucrase by Zymomonas mobilis

2005

Abstract The effect of some aliphatic (n-butanol to n-hexadecanol) and aromatic (benzyl and phenethyl alcohols), anesthetics (procaine) and surfactants (Tween 20 to Tween 80) on the secretion of levansucrase by the levan-producing strain of Gram-negative ethanologenic bacteria Zymomonas mobilis 113S were examined in this study. During incubation of Z. mobilis cells with sucrose (10 mM) a decrease of the levansucrase activity was observed in the presence of these amphiphilic compounds concomitantly with an increase of a total amount of protein in the medium. Since none of the compounds under study had any effect on enzyme activity in vitro observed structure- and concentration-dependent rela…

An interpretation of EPR spectra of azide ligated superoxide dismutase from Propionibacterium shermanii

1995

A self-consistent description of the EPR spectra of the azide ligated SOD is obtained by taking into account the general configuration of the crystal field splitting in the crystal field Hamiltonian. The spread in the rhombicity parameter due to the presence of different conformational substates is introduced.
