Tobacco cells contain a protein, immunologically related to the neutrophil small G protein Rac2 and involved in elicitor-induced oxidative burst.
1997
Abstract Suspension-cultured cells of Nicotiana tabacum generated active oxygen species (AOS) when they were treated with the proteinaceous elicitor, cryptogein. This response was blocked by diphenylene iodonium, an inhibitor of the neutrophil NADPH oxidase. When microsomal extracts of tobacco cells were probed with an antibody directed against the human small G protein Rac2, two immunoreactive proteins were detected at 18.5 and 20.5 kDa. The same experiment performed with cytosolic extracts of tobacco cells led to the observation of a strong immunoreactive protein at 21.5 kDa only in the cryptogein-treated cells. The appearance of this cytosolic protein was related to the production of AOS…
Acquired resistance triggered by elicitins in tobacco and other plants
1996
Elicitins are a family of proteins excreted byPhytophthora spp. They exhibit high sequence homology but large net charge differences. They induce necrosis in tobacco plants which then become resistant to the tobacco pathogenPhytophthora parasitica var.nicotianae. In stem-treated plants, resistance was not restricted to the site of elicitin application, but could be demonstrated by petiole inoculation at all levels on the stem. Resistance was already maximum after two days and lasted for at least two weeks. It was effective not only towardsP. p. var.nicotianae infection, but also against the unrelated pathogenSclerotinia sclerotiorum. In contrast to dichloroisonicotinic acid, an artificial i…
The combined action of 9 lipoxygenase and galactolipase is sufficient to bring about programmed cell death during tobacco hypersensitive response
2005
International audience; Oxylipins, derived from fatty acid hydroperoxides (FAHs), are thought to play different roles during plant pathogen interactions. During hypersensitive response (HR) some of them serve as signals necessary for defence gene activation whereas others could contribute to pathogen killing or could participate in the execution of plant programmed cell death (PCD) associated with this resistance. In order to address the role of these compounds in the latter process, we have closely observed lipid peroxidation, the first step of this metabolic pathway, under different situations which led either to accelerated or inhibited HR cell death. The oxidative process has been studi…
Type-2 histone deacetylases as new regulators of elicitor-induced cell death in plants
2011
voir Addenda, notes additionnelles complétant l'article : "Dahan, J., Hammoudi, V., Wendehenne, D., Bourque, S. (2011). Type 2 histone deacetylases play a major role in the control of elicitor-induced cell death in tobacco. Plant signaling & behavior, 6 (11), 1865-1867. DOI : 10.4161/psb.6.11.17848".; International audience; Plant resistance to pathogen attack is often associated with a localized programmed cell death called hypersensitive response (HR). How this cell death is controlled remains largely unknown. Upon treatment with cryptogein, an elicitor of tobacco defence and cell death, we identified NtHD2a and NtHD2b, two redundant isoforms of type-2 nuclear histone deacetylases (HDACs…
Type 2 histone deacetylases play a major role in the control of elicitor-induced cell death in tobacco

Type-2 histone deacetylases as new regulators of elicitorinduced cell death in plants. New Phytologist, 192 (1), 127–139. DOI: 10.1111/j.1469-8137.2011.03788.x; International audience; The cell death which characterizes the onset of the Hypersensitive Response (HR) is a very important weapon evolved by plants to block pathogen development. By the use of numerous plant/avirulent pathogen or plant/elicitor models, we have now obtained detailed signalling pathways allowing, after pathogen or elicitor perception, the control of the expression of specific sets of ge…
Preferential induction of 20S proteasome subunits during elicitation of plant defense reactions: towards the characterization of "plant defense prote…
2003
Plants have evolved efficient mechanisms to resist pathogens. The earliest defense response is the hypersensitive response (HR) considered as the main step leading to plant systemic acquired resistance (SAR) that protects the whole plant against a large spectrum of pathogens. We showed previously that elicitation of defense reactions in tobacco cells by cryptogein, a proteinaceous elicitor of plant defense reactions, leads to a rapid and differential accumulation of transcripts corresponding to genes encoding defense-induced (din) subunits of 20S proteasome: beta1din, alpha3din and alpha6din.Here, expression of these three subunits was investigated by Northern blotting and by Western blotti…
Hydrogen peroxide induces programmed cell death features in cultured tobacco BY-2 cells, in a dose-dependent manner
2001
Active oxygen species (AOS), especially hydrogen peroxide, play a critical role in the defence of plants against invading pathogens and in the hypersensitive response (HR). This is characterized by the induction of a massive production of AOS and the rapid appearance of necrotic lesions is considered as a programmed cell death (PCD) process during which a limited number of cells die at the site of infection. This work was aimed at investigating the mode of cell death observed in cultures of BY-2 tobacco cells exposed to H(2)O(2). It was shown that H(2)O(2) is able to induce various morphological cell death features in cultured tobacco BY-2 cells. The hallmarks of cell death observed with fl…
Involvement of putative glutamate receptors in plant defence signaling and NO production
2011
International audience; Ionotropic glutamate receptors (iGluRs) are non-selective cation channels permeable to calcium, present in animals and plants. In mammals, glutamate is a well-known neurotransmitter and recently has been recognized as an immunomodulator. As animals and plants share common mechanisms that govern innate immunity with calcium playing a key role in plant defence activation, we have checked the involvement of putative iGluRs in plant defence signaling. Using tobacco cells, we first provide evidence supporting the activity of iGluRs as calcium channels and their involvement in NO production as reported in animals. Thereafter, iGluRs were shown to be activated in response t…
Effect of ambient temperature variations on an indigenous microalgae-nitrifying bacteria culture dominated by Chlorella
2019
[EN] Two outdoor photobioreactors were operated to evaluate the effect of variable ambient temperature on an indigenous microalgae-nitrifying bacteria culture dominated by Chlorella. Four experiments were carried out in different seasons, maintaining the temperature-controlled PBR at around 25¿°C (by either heating or cooling), while the temperature in the non-temperature-controlled PBR was allowed to vary with the ambient conditions. Temperatures in the range of 15¿30¿°C had no significant effect on the microalgae cultivation performance. However, when the temperature rose to 30¿35¿°C microalgae viability was significantly reduced. Sudden temperature rises triggered AOB growth in the indig…
Different bacteria can enhance the micropropagation response of Cotoneaster lacteus (Rosaceae)
1998
C. MONIER, E. BOSSIS, C. CHABANET AND R. SAMSON. 1998. Cotoneaster species are widely used ornamental shrubs, and research is under way to select genotypes which are more resistant to fire blight by somaclonal variation. During the establishment of micropropagation, one genotype, Cotoneaster lacteus, showed the development of endogenous bacterial contaminants. Two strains were isolated from in vitro explants: Bacillus pumilus and Alcaligenes faecalis. These two strains, and some Pseudomonas fluorescens and Ps. putida, promoted in vitro multiplication and rooting of explants. In Pseudomonas strains, rooting enhancing capacity appeared to be related to indole-3-acetic acid (IAA) production, a…
