Expression and purification of polyhistidine-tagged firefly luciferase in insect cells

The coleopteran firefly, Photinus pyralis, luciferase was produced in lepidopteran Trichoplusia ni insect cells using a baculovirus expression vector. The recombinant protein was equipped with a polyhistidine affinity tag at the carboxyl terminus and purified by immobilized metal-ion affinity chromatography in combination with an expanded bed adsorption system. This approach enabled an efficient, one-step purification protocol of a genetically modified luciferase with properties similar to those of the authentic counterpart. Experimental Observation Of Turbulent Structures In A Straight Flume
2010
Experimental and theoretical studies of the structure of turbulence in open-channel flows have shown that the dynamics of the wall layer turbulence is dominated by the formation and growth of turbulent structures which evolve periodically as part of the so-called bursting phenomena. In the present paper experimental results obtained in a straight flume for different roughness conditions of the channel walls are described. The occurrence of turbulent events is verified by applying the conditioned quadrant analysis. The information about the spatial and temporal scales of the events is obtained through the space-time correlations of the conditioned data.

In this paper we propose a case study of urban heat island applied to Palermo. The urban heat island (UHI) is the most studied of the climate effects of settlements. The UHI refers to the generally warm urban temperatures compared to those over surrounding, non-urban, areas. The aim of this paper is to find a connection among the average rise in temperature and the modification of albedo.
The monophyly and evolution of Cynara L. (Asteraceae) sensu lato: evidence from the Internal Transcribed Spacer region of nrDNA

The monophyly and evolution of Cynara was investigated using ITS sequence data. Parsimony analysis supports the monophyly of Cynara sensu lato, i.e. including the distinctive taxa C. humilis and C. tournefortii. This contradicts the recent decision to create a new monotypic genus Arcyna for C. tournefortii. A hypothesised close relationship between C. tournefortii and Silybum Adans. is also refuted. Four of the five species of Cynara, for which multiple accessions were sequenced, were shown to be monophyletic but C. baetica was found to be non-monophyletic.
