Spectroscopic Methods for the Determination of Protein Interactions

2005

This unit provides guidelines on how to use steady-state fluorescence spectroscopy for the quantification of protein-protein interactions. The fluorescence of a protein is characterized by its excitation and emission spectra, quantum yield, and anisotropy. These parameters can change upon interaction with another protein and can be used to measure the extent of complex formation. The source of fluorescence can be an intrinsic fluorophore, such as tryptophan or tyrosine; a covalently attached fluorescent dye; or a fluorescent binding partner, such as a nucleotide or cofactor, that interacts specifically with the complex. Protocols are provided in this unit for determining affinity constants …

Heterometallic Titanium-Organic Frameworks as Dual Metal Catalysts for Synergistic Non-Buffered Hydrolysis of Nerve Agent Simulants

2020

Heterometallic metal-organic frameworks (MOFs) can offer important advantages over their homometallic counterparts to enable targeted modification of their adsorption, structural response, electronic structure, or chemical reactivity. However, controlling metal distribution in these solids still remains a challenge. The family of mesoporous titanium-organic frameworks, MUV-101(M), displays heterometallic TiM2 nodes assembled from direct reaction of Ti(IV) and M(II) salts. We use the degradation of nerve agent simulants to demonstrate that only TiFe2 nodes are capable of catalytic degradation in non-buffered conditions. By using an integrative experimental-computational approach, we rational…

On operads, bimodules and analytic functors

2017

We develop further the theory of operads and analytic functors. In particular, we introduce a bicategory that has operads as 0-cells, operad bimodules as 1-cells and operad bimodule maps as 2-cells, and prove that this bicategory is cartesian closed. In order to obtain this result, we extend the theory of distributors and the formal theory of monads.

HBIM data management in historical and archaeological buildings

2020

Recent technological evolutions in the acquisition and management of building data are offering new opportunities for digital reconstruction. At the same time, the BIM (Building Information Modeling) methodology, based on the implementation of libraries composed of parametric objects provided by the IFC (Industry Foundation Classes) standard, allows the design and management of data of existing buildings, and, in particular, historical and archaeological buildings. In the latter case, the great variety of Cultural Heritage (CH) distributed over the European territory, and the ability of BIM to cover the life of buildings or/and other artefacts from a geometric, descriptive, physical and sta…

Massless positivity in graviton exchange

2021

We formulate Positivity Bounds for scattering amplitudes including exchange of massless particles. We generalize the standard construction through dispersion relations to include the presence of a branch cut along the real axis in the complex plane for the Maldestam variable $s$. In general, validity of these bounds require the cancellation of divergences in the forward limit of the amplitude, proportional to $t^{-1}$ and $\log(t)$. We show that this is possible in the case of gravitons if one assumes a Regge behavior of the amplitude at high energies below the Planck scale, as previously suggested in the literature, and that the concrete UV behaviour of the amplitude is uniquely determined…

Development of the filiform hairs on the cerci of Gryllus bimaculatus Deg. (Saltatoria, Gryllidae)

1978

The filiform hairs, mechanoreceptors of Gryllus, pass through six developmental stages during the last larval stage. The cytoplasm of their sense cells suggests intensive synthesis of protein for cellular metabolism and intercytoplasmic exchange of material via glial evaginations. Ultrahistochemical tests demonstrated acid phosphatase in the lysosomes as well as in components of the Golgi apparatus. There was no significant change in the appearance of the sense cell cytoplasm, indicating a maintained functional state also during molting. The new cuticular apparatus is formed after apolysis by the three enveloping cells. Formation of the replacement hairs is initiated by a cytoplasmic outgro…

A new user interface for human-computer interaction in virtual reality environments

2010

This paper concerns the result of a research activity about the problems related to the human-computer interaction (both hardware and software) in virtual reality systems. In particular, a virtual reality application package, called VIRDE and used for the design review in immersive environment, has been used as a case study. After a detailed analysis of the hardware and software systems, and the examination of the main functional requirements, a new user interface has been developed. This new interface being composed of two input devices (tracked by an optical system) permits the two-hands interaction, instead of the most commonly single-hand interaction. The measurement of the software usa…

Potentialities of a Highway Alignment Optimization Method in an I-BIM Environment

2019

The BIM (Building Information Modeling) approach potential in the civil engineering field opened novel scenarios in the design idea concept, from planning to executive and constructive phases. The related advantages are numerous and not only limited to a real-time interaction among the involved subjects, that can actually operate in an optimized 3D shared environment. Owing to the sharing information philosophy and to the features of various "smart objects" combined in the project, this innovation reduces potential errors and increases the effectiveness of the design solution in terms of both functionality and cost. Despite these advantages, the highway alignment design problem remains very…
