DNA fragmentation index, pAKT and pERK1/2 in cumulus cells are related to oocyte competence in patients undergoing in vitro fertilization programme
2019
SummaryActivated pERK1/2 and pAKT are key players in supporting cell survival and proliferation pathways. Translocation of pERK1/2 into the nucleus, where it interacts with transcription factors and DNA itself, is instrumental in exerting an anti-apoptotic effect. In this study, pAKT levels, pERK1/2 nuclear localization and DNA fragmentation index (DFI) in cumulus cells of single cumulus–oocyte complexes of patients undergoing in vitro fertilization programmes were evaluated and correlated with the clinical outcome of the related embryos. For a positive clinical outcome of blastocyst development, pERK1/2 nuclear localization and DFI value had a significant inverse relationship, whereas the …
Dexrazoxane shows cytoprotective effects in zoledronic acid-treated human cells in vitro and in the rabbit tibia model in vivo
2012
Abstract Introduction Bisphosphonates are important and effective drugs in oncology and osteoporosis therapy. They accumulate in the bone matrix becoming released and active by bone resorption. This leads to effective inhibition of tumor cells and bone degradation. A side effect of bisphosphonates similar to other drugs like denosumab is osteonecrosis of the jaws (ONJ). This problem mostly occurs after tooth extraction. We studied the cytoprotectant dexrazoxane known from anthracycline chemotherapy for cytoprotection in nitrogen-containing bisphosphonate treated cells and in the rabbit tibia model to evaluate a possible value in ONJ management. Materials & methods Human osteoblasts (HOB) P2…
Paracrine effect of membrane vesicles released by mouse mesoangioblast stem cells
2016
Introduction: Mouse mesoangioblasts are vessel-associated multipotent progenitor stem cells, which are able to differentiate into different mesodermal cell types. In our previous paper, we have demonstrated that mesoangioblasts are able to shed in the extracellular environment membrane vesicles (EVs), which contain both structural proteins and biological factors such as FGF2 and the two gelatinases MMP2/9. We investigated whether these EV interact in a paracrine way with other cell types different from mesoangioblasts, and eventually the effects of this interaction. Methods: Mesoangioblast EVs were collected from conditioned media by ultracentrifugation. Total mRNAs from mesoangioblasts and…

Sulphate removal over barium-modified blast-furnace-slag geopolymer
2016
Blast-furnace slag and metakaolin were geopolymerised, modified with barium or treated with a combination of these methods in order to obtain an efficient SO4(2-) sorbent for mine water treatment. Of prepared materials, barium-modified blast-furnace slag geopolymer (Ba-BFS-GP) exhibited the highest SO4(2-) maximum sorption capacity (up to 119mgg(-1)) and it compared also favourably to materials reported in the literature. Therefore, Ba-BFS-GP was selected for further studies and the factors affecting to the sorption efficiency were assessed. Several isotherms were applied to describe the experimental results of Ba-BFS-GP and the Sips model showed the best fit. Kinetic studies showed that th…

The vesicular transfer of CLIC1 from glioblastoma to microvascular endothelial cells requires TRPM7.
2018
Chloride intracellular channel 1 (CLIC1) is highly expressed and secreted by human glioblastoma cells and cell lines such as U87, initiating cell migration and tumor growth. Here, we examined whether CLIC1 could be transferred to human primary microvascular endothelial cells (HMEC). We previously reported that the oncogenic microRNA, miR-5096, increased the release of extracellular vesicles (EVs) by which it increased its own transfer from U87 to surrounding cells. Thus, we also examined its effect on the CLIC1 transfer. In homotypic cultures, miR-5096 did not increase the expression of CLIC1 in U87 nor in HMEC. However, the endothelial CLIC1 level increased after exposure to EVs released b…
Does the evidence matter in medicine? The retinoblastoma paradigm.
2007
Retinoblastoma (Rb) is the most common intraocular malignant tumour in childhood, with an incidence of 1 in 15,000 live births. Complete information on this rare tumour can be easily accessed through the internet, although many aspect concerning the aetiology and pathogenesis of the disease, are still controversial. The "two hit" theory, formulated in 1971 to explain the variegated clinical expression of the disease, is based on the idea that single gene mutation may determine the development of cancer. However, this view does not take into account the most recent evidences showing the role of aneuploidy and chromosome instability in cancer. Also, a number of other genes and epigenetic mech…
