Some Plant Defense Stimulators can induce IL-1β production in human immune cells in vitro

2020

8 pages; International audience; Among Plant Protection Products (PPP), a new emerging category of pesticides act by stimulating plant defense in order to improve plant resistance against microbial pathogens. Given that these compounds, the so-called Plant Defense Stimulators (PDS) act on innate immunity, we tested, using an in vitro approach on human mononuclear leucocytes (PBMC), the potential toxicity (XTT assay) and inflammatory effects (production of IL-1β) of 4 PPP belonging to different chemical families. We found that two products (LBG-01F34® and Regalis®) did not induce any cytotoxicity or IL-1 β production. The product BION-50 WG®, that contains Acibenzolar-S-methyl (ASM) and sili…

Effet anti-tumoral de l'acide docosahexaénoïque : implication des microARNs et du TNFalpha

2015

Docosahexaenoic acid (DHA) is an omega-3 polyunsaturated fatty acid with anti-inflammatory and anti-tumoral properties. The anti-tumor effect of DHA in colorectal cancer might be attributed to direct anti-proliferative action on cancer cells and to its ability to reduce inflammatory status involved in tumor growth. Tumor Necrosis Factor-alpha (TNFa) is an inflammatory cytokine with paradoxical effect in cancer biology. According to the cellular context, TNFa activates RIP1 kinase dependent signaling pathway leading to proliferation or cell death. Our aim was to evaluate the role of TNFa in anti-proliferative effect of DHA in colon cancer cells and to precise the molecular mechanisms regulat…

L'hyperthermie provoque l'agrégation de FLIP et restaure l'apoptose induite par TRAIL

2014

The TNF-family member TRAIL (TNF-related apoptosis inducing ligand) is a cytokine involved in the immune anti-tumour surveillance. TRAIL is a promising agent currently under investigation for its anti-cancer properties with limited side effects on healthy cells. However, the use of TRAIL in oncology has been limited due to its lack of efficiency, mainly associated with cell resistance to apoptosis. The aim of this project was to study the interest of hyperthermia (or heat shock - HS) as an adjuvant for TRAIL therapy and the mechanisms involved in this sensitization. We have first evaluated the significance of this combination in a large variety of cancer cell lines known to be sensitive or r…

Analysis of E2F1 by clAP1

2017

The cellular inhibitor of Apoptosis 1 (cIAP1) behaves as an E3 ubiquitin ligase and has oncogenic properties. Previously, our team has shown that cIAP1 can regulate the E2F1 transcription factor activity. My research project has been focused on deepening our current knowledge on this interaction. Firstly, we characterized the E2F1-cIAP1 interaction, then we analyzed the regulation of E2F1 by cIAP1 and finally assessed the importance of the cIAP1-E2F1 interaction for the oncogenic properties of cIAP1. I have demonstrated a interaction of E2F1 with the hydrophobic pocket of the BIR3 domain of cIAP1. Moreover, I highlighted that the alpha 1 helix of the BIR3 domain is mandatory for the stabili…

Control of the HOXA9 gene expression in the hematopoietic stem/progenitor cells : role of the epigenetic factors MOZ, MLL and of the polyadenylation …

2013

My thesis project has consisted of the study of MOZ, and MLL. They are epigenetic regulators. MOZ and MLL activate transcription of HOX genes, which are transcription factors essential during haematopoiesis. MOZ and MLL have some target genes in common. In our study, we characterised a cooperation between MOZ and MLL in human haematopoietic stem/progenitor cells CD34+. They are both recruited onto HOX promoters. MOZ is essential for MLL recruitment, and this is reciprocal. In conclusion, we provided an example of a mechanism involving a direct cross-talk between two histone modifying enzymes. In order to dissect the mechanism of action of this complex, we decided to identify novel proteins i…

Analysis of eIF4E gene sconfering resistance to Pea seed-borne mosaic virus (PSbMV) and other potyviruses in pea

2013

Pea seed-borne mosaic virus (PSbMV) belongs among the most frequent viral pathogens causing severe losses in field pea and other legumes. These losses might be prevented by growth of resistant varieties. Resistance to the common P1 strain of PSbMV isconferred by a single recessive gene (eIF4E), localized on LG VI (sbm-1 locus), while to lentil L1 strainis localized on LGII atsbm-2locus. Gene-specific single nucleotide polymorphism and co-dominant amplicon length polymorphism markers were developed. From the comparison of genotype verses phenotype for selection of resistance/susceptibility in F2 plants, there was a 26% discrepancy between the PCR and ELISA-based assays with potentially susce…
