Bone metastases have a major impact on morbidity and on mortality in cancer patients. Despite its clinical relevance, metastasis remains the most poorly elucidated aspect of carcinogenesis. The biological mechanisms leading to bone metastasis establishment have been referred as " vicious circle," a complex network between cancer cells and the bone microenvironment. ABSTRACT Background Data are limited regarding bone metastases from colorectal cancer (CRC). The objective of this study was to survey the natural history of bone metastasis in CRC. Patients and methods This retrospective, multicenter, observational study of 264 patients with CRC involving bone examined cancer treatments, bone metastases characteristics, skeletal-related event (SRE) type and frequency, zoledronic acid therapy, and disease outcomes. Results Most patients with bone metastases had pathologic T3/4 disease at CRC diagnosis. Preliminary Results of Two Eortc Randomized Trials in Previously Untreated Patients with Advanced T3 — T4 Prostatic Cancer

At the end of 1976 and the beginning of 1977 respectively, the EORTC Genito-Urinary Tract Cancer Cooperative Group started two randomized phase III protocols in previously untreated patients with advanced T3–4 prostatic cancer (1–3). The first trial, 30762, compared Stilboestrol and Estracyt, while the second, 30761, was a randomized comparison of Stilboestrol, Cyproterone Acetate and Medroxyprogesterone Acetate. Both trials have now been closed to patient entry and the principal endpoints of the studies analyzed. This paper will present the interim results which have emerged from these two studies.

Metalloproteinases (MMP)-13 is expressed in a broad range of primary malignant tumours and it is emerging as a novel biomarker. Recent data suggest a direct role of MMP-13 in dissolving bone matrix complementing the activity of MMP-9 and other enzymes. Bone metastasis develops in 30% of all patients with renal cell carcinoma. We elucidated the mechanisms that lead to and predict bone metastasis of renal cell carcinoma.Nine renal cell carcinoma primary cell lines and 30 renal cell carcinoma tissue specimens (normal and tumor tissue) were collected from 3 patients with no metastasis and 10 with lung or bone metastasis within 5 years after nephrectomy. Activin A, is a multifunctional cytokine of the transforming growth factor-b superfamily of growth factors. This molecule has been shown to be implicated in the regulation of a broad range of important biological functions including bone remodelling. Therefore, a deregulation in the activin signalling pathway may result in disturbances of normal bone metabolism and, eventually, in the onset of severe pathological conditions associated with an altered bone resorption.
