The role of oxygen in tumor cell proliferation, radiosensitivity, cytotoxicity of anticancer drugs and hyperthermia treatment has been the subject of a series of investigations (for reviews see Hall, 1988; Teicher et al., 1990, Vaupel et al., 1989a; Vaupel, 1990a,b). Despite the apparent importance of tumor oxygenation, data on pO2 values in solid tumors are mostly derived from experiments on rodents which might not necessarily reflect the variability of the clinical situation. Adjuvant denosumab in early breast-cancer

We read with interest the D-CARE trial report by Robert Coleman and colleagues1 on the adjuvant administration of denosumab in patients with early-stage breast cancer. Despite the negative results, which pose concerns about high dose denosumab schedules in an adjuvant setting, this large and well- designed trial can provide useful clinical data that are usually difficult to obtain from observational studies, particularly regarding medication- related osteonecrosis of the jaw (MRONJ).

Triple-negative breast cancers (TNBCs) are a heterogenous group of breast cancers characterized by poor prognosis because they are not amenable to targeted therapies. We have taken into account that altered expression of Raf-1 kinase inhibitor protein (RKIP), a tumor and metastasis suppressor and a promoter of drug-induced apoptosis, is frequent in TNBCs and may be involved in their aggressive biology. Interestingly, the analysis of the possible mechanisms of RKIP downregulation in TNBCs permits the identification and recapitulation of different possible approaches, including epigenetic modulation, e.g., by DNA demethylating agents or histone deacetylase inhibition, and NF-κB inhibition.
