Defining substance use disorders: do we really need more than heavy use?

Aims: The aim of the study was to explore whether the concept of heavy substance use over time can be used as definition of substance use disorder. Methods: Narrative review. Results: Heavy use over time clearly underlies the neurobiological changes asso- ciated with current thinking of substance use disorders. In addition, there is evidence that heavy use over time can explain the majority of social problems and of burden of disease (morbidity and mortality). The phenotype of NFATc2(-/-) c3(-/-) (double knockout [DKO]) mice implies a disturbed regulation of T cell responses, evidenced by massive lymphadenopathy, splenomegaly, and autoaggressive phenomena. The population of CD4(+) CD25(+) T cells from DKO mice lacks regulatory capacity, except a small subpopulation that highly expresses glucocorticoid-induced tumor necrosis factor receptor family-related gene (GITR) and CD25. However, neither wild-type nor DKO CD4(+) CD25(+) regulatory T cells (T reg cells) are able to suppress proliferation of DKO CD4(+) CD25(-) T helper cells. Insect flight is one of the most intense and energy-demanding physiological activities. High carbohydrate oxidation rates are necessary for take-off, but, to spare the limited carbohydrate reserves, long-distance flyers, such as locusts, soon switch to lipid as the main fuel. We demonstrate that before a flight, locust muscles are metabolically poised for take-off by the release of octopamine from central modulatory dorsal unpaired median (DUM) neurons, which increases the levels of the potent glycolytic activator fructose 2,6-bisphosphate in flight muscle. Context specificity of both acquisition and extinction of a Pavlovian conditioned response

It is widely held that the extinction of a conditioned response is more context specific than its initial acquisition. One proposed explanation is that context serves to disambiguate the meaning of a stimulus. Using a procedure that equated the learning histories of the contexts, we show that the memory of an appetitive Pavlovian association can be highly context specific despite being unambiguous. This result is inconsistent with predictions of the Rescorla–Wagner model of learning but in line with configural accounts of contextual control of behavior. Students of Religion Studying Social Conflict Through Simulation and Modelling: An Exploration

Researchers at our university use modelling and simulation (M&S) to study religious conflicts, and we wanted to introduce undergraduate students of religion to this research approach. Hence, we started a three-year educational design research project to empirically study ways to introduce these students to M&S as a viable research method in their discipline. The research project will entail several iterations, which aim to have a feasible and effective design of lessons and a better understanding of the learning processes. A method for measuring mitochondrial mass and activity

Mitochondria, responsible for the energy-generating process essential for the cell metabolism, differ for the number, localization and activity in animal cells and tissues in relation to the energetic needs. Using fluorescent probes specific for mitochondria, Mitotracker Green (MTG) and Orange (MTO), and Confocal Laser-Scanning Microscope (CLSM), we elaborated a method to measure in vivo the mitochondrial mass and activity, in sea urchin Paracentrotus lividus eggs and embryos. The analysis of captured images, revealed a variation of mitochondrial distribution and an increase of activity after fertilization.
