Regulation of stress response in Oenococcus oeni as a function of environmental changes and growth phase

International audience; Oenococcus oeni is a lactic acid bacterium which is able to grow in wine and perform malolactic fermentation. To survive and grow in such a harsh environment as wine, O. oeni uses several mechanisms of resistance including stress protein synthesis. The molecular characterisation of three stress genes hsp18, clpX, trxA encoding for a small heat shock protein, an ATPase regulation component of ClpP protease and a thioredoxin, respectively, allow us to suggest the existence in O. oeni of multiple regulation mechanisms as is the case in Bacillus subtilis. A Trans-amplifying RNA Vaccine Strategy for Induction of Potent Protective Immunity

Here, we present a potent RNA vaccine approach based on a novel bipartite vector system using trans-amplifying RNA (taRNA). The vector cassette encoding the vaccine antigen originates from an alphaviral self-amplifying RNA (saRNA), from which the replicase was deleted to form a transreplicon. Replicase activity is provided in trans by a second molecule, either by a standard saRNA or an optimized non-replicating mRNA (nrRNA). The latter delivered 10- to 100-fold higher transreplicon expression than the former. ABSTRACT The random transposon mutagenesis system P junc -TpaseIS 1223 is composed of plasmids pVI129, expressing IS 1223 transposase, and pVI110, a suicide transposon plasmid carrying the P junc sequence, the substrate of the IS 1223 transposase. This system is particularly efficient in Lactobacillus casei , as more than 10,000 stable, random mutants were routinely obtained via electroporation.

Soil erosion assessment on tillage and alternative soil managements in a Sicilian vineyard

Mediterranean crops favour high erosion rates. Vineyards use to reach the highest soil and water losses due to the lack of vegetation cover. A topographical approach by means of the use of vineyards poles as fixed reference point as erosion markers allowed to quantify high and non-sustainable soil erosion rates on the Sicilian vineyards during 9 years. In order to develop strategies to control the soil losses, seven land management were selected and applied in a typical blanc wine grape vineyard located in southwestern Sicily. Capturing colorectal cancer inter-tumor heterogeneity in patient-derived xenograft (PDX) models

Patient‐derived xenograft (PDX) models have become an important asset in translational cancer research. However, to provide a robust preclinical platform, PDXs need to accommodate the tumor heterogeneity that is observed in patients. Colorectal cancer (CRC) can be stratified into four consensus molecular subtypes (CMS) with distinct biological and clinical features. Surprisingly, using a set of CRC patients, we revealed the partial representation of tumor heterogeneity in PDX models.
