Genome-Wide Association Analysis in Primary Sclerosing Cholangitis

Background & Aims We aimed to characterize the genetic susceptibility to primary sclerosing cholangitis (PSC) by means of a genome-wide association analysis of single nucleotide polymorphism (SNP) markers. Methods A total of 443,816 SNPs on the Affymetrix SNP Array 5.0 (Affymetrix, Santa Clara, CA) were genotyped in 285 Norwegian PSC patients and 298 healthy controls. Associations detected in this discovery panel were re-examined in independent case-control panels from Scandinavia (137 PSC cases and 368 controls), Belgium/The Netherlands (229 PSC cases and 735 controls), and Germany (400 cases and 1832 controls). The paper analyzes the achievable strategic role that the porto of Porto Empedocle (sinergy by Agrigento, its "city mother") could have. This marginal city, localized in the center of the African side of the Sicily, is far from European politics, but this situation could change. It's necessary to improve the sea routes and to activate the disused railway line from Porto Empedocle to Castelvetrano (near Trapani). Laser speckle contrast analysis (LASCA) offers a non-contact, full-field, and real-time mapping of capillary blood flow and can be considered as an alternative method to Laser Doppler perfusion imaging. In vivo confocal laser laparoscopy allows real time subsurface microscopy in animal models of liver disease.

Background/Aims Histopathology is essential in the diagnostic workup of most liver diseases. However, biopsy sampling might carry risks, is subject to sampling error, and does not provide dynamic tissue imaging. Therefore a newly developed miniaturised confocal probe was evaluated for in vivo microscopic imaging in rodent models of human liver diseases. Methods The handheld laparoscopy probe used a 488nm single line laser for fluorophore excitation. Optical slice thickness was 7μm, lateral resolution 0.7μm. The flow of bile secretion into the human digestive system was simulated by the dilution of a bile salt-lipid micellar solution. The structural development upon the dilution of the fed state bile model FeSSIF(mod6.5) to the fasted state bile model FaSSIF(mod) was investigated by small-angle neutron scattering (SANS) and dynamic light scattering (DLS) in crossed beam experiments to observe small and large structures in a size range of 1 nm to 50 μm in parallel. A Lipopolysaccharide Antigen of the Treponema

IMMUNOCHEMICAL studies carried out by D'Alessandro et al. 1 on a non-pathogenic, culturable treponeme, the so-called Reiter treponeme, revealed the presence of four antigenic components: (1) a thermolabile protein; (2) a specific, thermostable antigen with polysaccharide characteristics; (3) a lipoid, corresponding to the ubiquitous lipidic antigen cardiolipin; (4) another lipoid similar to the organ-specific cerebral antigen of Witebsky2. These studies led to the conclusion that the treponemes, like other organisms, are mosaics of antigens, and a safe basis was established for a better understanding of the complex serological response of the infected host.

Plasma PLTP (phospholipid-transfer protein) is a member of the lipid transfer/LBP [LPS (lipopolysaccharide)-binding protein] family, which constitutes a superfamily of genes together with the short and long PLUNC (palate, lung and nasal epithelium clone) proteins. Although PLTP was studied initially for its involvement in the metabolism of HDL (high-density lipoproteins) and reverse cholesterol transport (i.e. the metabolic pathway through which cholesterol excess can be transported from peripheral tissues back to the liver for excretion in the bile), it displays a number of additional biological properties.
