X-linked adrenoleukodystrophy (X-ALD) is a complex neurodegenerative disease associated with mutations in the ABCD1 gene, which encodes for a peroxisomal ABC transporter. Thanks to the efforts of the ELA foundation and to the recent successes of gene therapy published in Science in 2009, X-ALD is better known but still remains poorly understood. The exact role of ABCD1 and its homologs, as well as the exact link between the biochemical and metabolic peroxisomal defects and the clinical symptoms of the disease remain to be elucidated. This review summarizes the knowledge concerning the subfamily D of the ABC transporter family and concerning X-ALD, the most frequent peroxisomal disorder.

Interpatient variability in drug response can be widely explained by genetically determined differences in metabolizing enzymes, drug transporters, and drug targets, leading to different pharmacokinetic and/or pharmacodynamic behaviors of drugs. All polymeric chemosensitizers proposed thus far have a linear poly(ethylene glycol) (PEG) hydrophilic block. To testify whether precisely this chemical structure and architecture of the hydrophilic block is a prerequisite for chemosensitization, we tested a series of novel block copolymers containing a hyperbranched polyglycerol segment as a hydrophilic block (PPO-NG copolymers) on multi-drug-resistant (MDR) tumor cells in culture. The search for novel SUR1-ligands originates from the idea to influence the in vivo behaviour by adding new structural moieties to the glibenclamide structure while preserving its binding affinity. Observations of the SNR Cassiopeia A (Cas A) show asymmetries in the reverse shock that cannot be explained by models describing a remnant expanding through a spherically symmetric wind of the progenitor star. We investigate whether a past interaction of Cas A with an asymmetric circumstellar shell can account for the observed asymmetries. We performed 3D MHD simulations that describe the remnant evolution from the SN to its interaction with a circumstellar shell. We discuss gauge theory with a topological N=2 symmetry. This theory captures the de Rham complex and Riemannian geometry of some underlying moduli space $\cal M$ and the partition function equals the Euler number of $\cal M$. We explicitly deal with moduli spaces of instantons and of flat connections in two and three dimensions. To motivate our constructions we explain the relation between the Mathai-Quillen formalism and supersymmetric quantum mechanics and introduce a new kind of supersymmetric quantum mechanics based on the Gauss-Codazzi equations.
