Electrochemical Characterization of Archaeological Tin-Opacified Lead-Alkali Glazes and Their Corrosion Processes

The electrochemical response of weathered and unweathered archaeological tin-opacified glazes attached to paraffin-impregnated graphite electrodes is described. Upon comparison with the square wave voltammetric response of SnO2, PbO and PbO2, Sn- and Pb-centered reduction processes can be characterized. Reduction of Sn(IV) involves the stepwise formation of solid Sn(II) and Sn metal, successively, at potentials of −0.08 and −0.55 V vs. AgCl/Ag. Reduction of network-modifier Pb(II) in glazes occurs at −0.5 V and is accompanied by the reduction of network-forming Pb(IV) at potentials ranging from +0.65 to +0.20 V, confirming the presence of such centers in glazes. Enzymatic Synthesis and Surface Deposition of Tin Dioxide using Silicatein-α

Nanostructured tin dioxide was synthesized by making use of the catalytic activity of silicatein-α. TEM, HRTEM, and XRD revealed the formation of cassiterite SnO2. Surface bound silicatein retains its biocatalytic activity. This was demonstrated by immobilizing silicatein on glass surfaces using a histidine-tag chelating anchor. The subsequent deposition of SnO2 on glass was monitored by quartz crystal microbalance (QCM) measurements and scanning electron microscopy (SEM). This new aspect of silicatein activity toward the formation of metal oxides other than SiO2, TiO2, and BaTiO3 opens up new vistas in composite material synthesis.

Real-Time RT-PCR assay to quantify the expression of fum1 and fum19 genes from the Fumonisin-producing Fusarium verticillioides

Fumonisins are a group of mycotoxins produced by Fusarium species of the Gibberella fujikuroi species complex that contaminate food and feed products, and represent a risk for human and animal health. In this work, we have developed a specific real-time reverse transcription-PCR (RT-PCR) assay to quantify the level of expression of two genes of the fumonisin biosynthetic cluster in F. verticillioides: fum1 (that encodes a polyketide synthase enzyme) and the ABC transporter encoding gene fum19. Methods An IncP1 plasmid from Achromobacter xylosoxidans carrying the cassette array dfrA14–blaVEB-1–aadB was introduced by conjugation into five strains of P. mirabilis: PmBRI, PmABB, PmSCO and Pm2CHAMA harbouring Salmonella GI 1 and PmESC harbouring Proteus GI 1.
