Abstract Harlequin ichthyosis (HI) is a severe autosomal recessive developmental disorder of the skin that is frequently but not always fatal in the first few days of life. In HI, mutations in both ABCA12 gene alleles must have a severe impact on protein function and most mutations are truncating. The presence of at least one nontruncating mutation (predicting a residual protein function) usually causes a less severe congenital ichthyosis (lamellar ichthyosis or congenital ichthyosiform erythroderma). The complete process of gene therapy involves three important steps: targeting, delivery, and gene expression. Since each step can be related to the pharmacological concept of affinity, bioavailability, and intrinsic capacity, this commentary examines, from this perspective, the efficiency of anionic and cationic liposomes as vectors for the in vivo gene transfer of the human alpha1-antitrypsin gene. Small liposomes represent the first generation of liposomes destined for the liver parenchymal cell. Cordycepin is one of the substantial components of the parasitic fungus Cordyceps sinensis as well as other Cordyceps species. It exerts various effects such as antimetastatic, antiinflammatory, antioxidant, and neuroprotective activities. Assorted studies revealed in vitro and in vivo anticancer influence of cordycepin and put forward its potential for cancer therapy. Multidrug resistance is a prevailing phenomenon leading to chemotherapy treatment failure in cancer patients. In the current study two known cytotoxic pseudoguaianolide sesquiterpene lactones; neoambrosin (1) and damsin (2) that circumvent MDR were identified. The two cytotoxic compounds were isolated using column chromatography, characterized using 1D and 2D NMR, MS, and compared with literature values. The isolated compounds were investigated for their cytotoxic potential using resazurin assays and thereafter confirmed with immunoblotting and in silico studies. P-glycoprotein belongs to the family of ATP-binding cassette (ABC) transporters. It functions in cellular detoxification, pumping a wide range of xenobiotic compounds, including anticancer drugs out of the cell. In cancerous cells, P-glycoprotein confers resistance to a broad spectrum of anticancer agents, a phenomenon termed multidrug resistance. An attractive strategy for overcoming multidrug resistance is to block the transport function of P-glycoprotein and thus increase intracellular concentrations of anticancer drugs to lethal levels. ABSTRACT The two-component system MtrBA is involved in the osmostress response of Corynebacterium glutamicum . MtrB was reconstituted in a functionally active form in liposomes and showed autophosphorylation and phosphatase activity. In proteoliposomes, MtrB activity was stimulated by monovalent cations used by many osmosensors for the detection of hypertonicity. Although MtrB was activated by monovalent cations, they lead in vitro to a general stabilization of histidine kinases and do not represent the stimulus for MtrB to sense hyperosmotic stress.
Optical and near-infrared recombination lines of oxygen ions from Cassiopeia A knots

Context. Fast-moving knots (FMK) in the Galactic supernova remnant Cassiopeia A consist mainly of metals and allow to study element production in supernovae and shock physics in great detail. Aims. We work out theoretically and suggest to observe previously unexplored class of spectral lines -- metal recombination lines in optical and near-infrared bands -- emitted by the cold ionized and cooling plasma in the fast-moving knots. Fine-structure infrared lines from the Cassiopeia A knots

Aims: Archival observations of infrared fine-structure lines of the young Galactic supernova remnant Cassiopeia A allow us to test existing models and determine the physical parameters of various regions of the fast-moving knots (FMKs), which are metal-dominated clouds of material ejected by the supernova explosion. Methods: The fluxes of the far-infrared [O i] and [O iii] lines are extracted from the previously unpublished archival ISO data. The archival Spitzer data are used to determine the fluxes of the O, Ne, Si, S, Ar and Fe ion fine-structure lines originating in the FMKs.
