An experiment was carried out on 12 Douglas fir seedlings lots that were 3 years old and had all originated from the same seed lot. Treatments consisted in combining stock type with three different height to diameter ratios, storage duration and method (long at 2°C or short in various conditions), and protection from desiccation (by bagging or not). Seedling lots were assessed at planting by root electrolyte leakage (REL), root moisture content (RMC) and predawn shoot water potential (Ywp). They were planted simultaneously in well-watered or water-stressed conditions. Performance level was based on survival and height growth at the end of the growing season. The high resolution of Landsat Thematic Mapper (TM) thermal infrared data can be interesting in applications such as energy balance and crop water stress studies. However, surface temperature measurements are strongly affected by atmospheric effects, particularly by water vapour absorption, therefore a correction method is absolutely necessary. The impossibility of applying a split-window equation and the difficulty of having temporarily coincident radiosondes produce a slight use of Landsat TM data to obtain surface temperature. Molecular-based methods are sometimes not sensitive enough to detect low-titre phytoplasmas in woody hosts. In order to enable the reliable routine diagnosis of Grapevine Yellows in dormant grapevine material, INRA-Dijon constructed new FD9 primers which, used in one step-PCR, proved very effective for the detection of phytoplasmas in symptomatic grapevine and elm. Used in nested-PCR assays, they provided an excellent signal from phloem chips or whole tissues taken from either canes or roots of dormant plants.
