Centrifuge tests on strip footings on sand with a weak layer
2017
Tests on small-scale physical models of a strip footing resting on a dense sand bed containing a thin horizontal weak soil layer were carried out at normal gravity (1 g ). The results, reported in a companion paper, point out that the weak layer plays an important role in the failure mechanism and the ultimate bearing capacity of the footing if it falls within the ground volume relevant to the behaviour of the sand–footing system. The same problem was also investigated by means of centrifuge tests on reduced-scale models at 25 g and 40 g . The results of these tests, reported and discussed in this paper, confirm that failure mechanisms are governed substantially by the presence of the weak…
Enhanced oxidative susceptibility and reduced antioxidant content of metabolic precursors of small, dense low-density lipoproteins.
2001
Abstract PURPOSE: Elevated plasma concentrations of low-density lipoproteins (LDL) increase risk for coronary heart disease. However, lipoprotein profiles rich in small, dense LDL particles confer greater risk than those that mainly consist of large, buoyant LDL. This may be due, in part, to the greater oxidative susceptibility of small, dense LDL. In the current studies, we tested whether differences in the oxidative behavior of buoyant and dense LDL arise from differences in their immediate metabolic precursors, intermediate-density lipoproteins. SUBJECTS AND METHODS: We compared the properties of intermediate-density lipoproteins and buoyant and dense LDL subfractions in 9 subjects with …
Vestibular Stimulation Interferes with the Dynamics of An Internal Representation of Gravity
2016
The remembered vanishing location of a moving target has been found to be displaced downward in the direction of gravity ( representational gravity) and more so with increasing retention intervals, suggesting that the visual spatial updating recruits an internal model of gravity. Despite being consistently linked with gravity, few inquiries have been made about the role of vestibular information in these trends. Previous experiments with static tilting of observers’ bodies suggest that under conflicting cues between the idiotropic vector and vestibular signals, the dynamic drift in memory is reduced to a constant displacement along the body's main axis. The present experiment aims to replic…
Numerical analysis of density gradient centrifugation profiles from eukaryotic DNA
1990
A numerical method for the deconvolution of superimposed Gaussian distributions with a unique solution has been proposed by Medgyessy [10]. We have tested the usefulness of this method for the analysis of density gradient centrifugation profiles from eukaryotic DNA, which are normally composed from overlapping Gaussian distributed profiles of several subcomponents with different mean buoyant densities. From the analysis of human DNA and from model calculations we conclude that major subcomponents can be identified by this method, if they differ in their buoyant density by approximatly 0.005 g/ml. Minor components can only be identified if the total DNA has been fractionated according to buo…
Ein schnelles on-line trennsystem unter anwendung von helium-jet- und zentrifugentechnik
1975
Abstract A centrifuge for continuous liquid-liquid phase separation is described. It is designed for rapid solvent extraction of short-lived radioactive isotopes (half-lives in the order of seconds). The centrifuge is characterized by simple construction and operation principle, easy handling, low cost, and a minimum hold-up time of less than five seconds (phase purity 99.0–99.9%). The on-line connection to a helium-jet transport system is described. Problems concerning the transfer of activities into a liquid phase from the helium-jet at atmospheric pressure are discussed.
Molecular basis of a new type of C1q-deficiency associated with a non-functional low molecular weight (LMW) C1q: parallels and differences to other k…
1998
Analysis of an abnormal C1q molecule of individuals of a Moroccan family by ultracentrifugation in sucrose gradients revealed a low molecular weight C1q (LMW-C1q). We investigated the molecular basis of this defect by sequencing all six exons of the three C1q genes. One point mutation in the codon for Gly at position 15 (GGT) of the B chain was found resulting in an amino acid substitution to Asp (GAT). The exchange not only leads to an interruption of the collagen-like motif Gly-X-Y, but also introduces one negatively charged residue per B chain which results in two additional charges per structural subunit (A-B, C-C, A-B). The mutation which has been identified by DNA-sequencing in the C1…
Isolation and Analysis of Fasciola hepatica Extracellular Vesicles
2020
The finding of extracellular vesicles (EVs) as important players in parasite-parasite and host-parasite communications has led to an increasing number of reports in the literature. Different protocols have been developed for isolation and further characterization of EVs from parasitic helminths. In this chapter, we describe step by step procedures to isolate EVs secreted by Fasciola hepatica adults in culture, which could be also applied for other developmental stages of the parasite, as well as EVs present in plasma and urine. Along with classical isolation methods like differential ultracentrifugation, and more recent techniques like size exclusion chromatography (SEC), here we also refer…
Separation of chitosomal chitin synthetase from cell-free extracts ofNeurospora crassa “Slime” variant agglutinated with concanavalin A
1989
Cell-free extracts of the wall-less slime variant ofNeurospora crassa were treated with concanavalin A (Con A); this treatment caused a massive agglutination of the particulate structures in the cell-free homogenate, although most (73%) of the chitin synthetase initially present in the cell-free extract remained in the supernatant obtained after sedimentation of the lectin-flocculated material. This chitin synthetase showed the sedimentation properties of chitosomes (unique microvesicular structures) and failed to bind [3H]Con A. A significant percentage (42%) of the chitin synthetase activity associated with the Con A-flocculated material probably corresponds to mechanically trapped chitos…
Field-free two-direction alignment alternation of linear molecules by elliptic laser pulses
2005
We show that a linear molecule subjected to a short specific elliptically polarized laser field yields postpulse revivals exhibiting alignment alternatively located along the orthogonal axis and the major axis of the ellipse. The effect is experimentally demonstrated by measuring the optical Kerr effect along two different axes. The conditions ensuring an optimal field-free alternation of high alignments along both directions are derived.
Pharmakologisch-aktive polymere, 21. Orientierende Untersuchungen zur körperverteilung und Ausscheidung von poly[2-(methylsulfinyl)ethylacrylat]en be…
1980
Poly[2-(methylsulfinyl)ethyl acrylate] (1) was synthesized as well as derivatives 14C-labelled in side groups (6) or 14C-labelled in the main chain (11). Polymer 11 with the 14C-labelled main chain was fractionated by precipitation. The η-M-relation determined by measurements of unlabelled polymers in the ultracentrifuge for comparison was used to establish the viscosimetrically determined molecular weights of the labelled fractions. After intravenous application of aqueous solutions of the polymer in rats the excretion rate up to 72 h after treatment was ascertained to ca. 50%; the concentration in the blood serum was found to be strikingly high. A tendency to reinforced storage in organs …
