Medium-size droplets of methyl ricinoleate are reduced by cell-surface activity in the gamma-decalactone production by Yarrowia lipolytica.

International audience; Size of methyl ricinoleate droplets during biotransformation into gamma-decalactone by Yarrowia lipolytica was measured in both homogenized and non-homogenized media. In non-homogenized but shaken medium, droplets had an average volume surface diameter d32 of 2.5 microm whereas it was 0.7 microm in homogenized and shaken medium. But as soon as yeast cells were inoculated, both diameters became similar at about 0.7 microm and did not vary significantly until the end of the culture. The growth of Y. lipolytica in both media was very similar except for the lag phase which was lowered in homogenized medium conditions.

Lipid composition of the vacuolar membrane of Acer pseudoplatanus cultured cells

Tonoplast was prepared by osmotic lysis of a pure vacuolar fraction isolated from protoplasts derived from Acer pseudoplatanus cultured cells. After their extraction, neutral and polar lipids were separated by a thin layer chromatography. Phospholipids, glycolipids and neutral lipids represented 44.5%, 39.1% and 16.4% of total lipids, respectively. Sterols (glycosylated plus non-glycosylated forms) constituted 30.8% of total lipids; 75% of sterols were glycosylated. The most prominent lipids were phosphatidylethanolamine (20.8%), phosphatidylcholine (13.5%), ceramide monohexoside (12.8%), steryl glycoside (12.2%) and acylated steryl glycoside (10.9%). Abstract The thermochemical behavior of various samples of okra ( Abelmoschus esculentus ) and miscanthus ( Miscanthus x giganteus ) stalks (initial, hot-water-extracted, and those from sulfur-free delignification) were studied by pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS). In all cases, major GC-amenable condensable products were measured semi-quantitatively and classified into several product groups. Effect of Enzyme Amounts Used in Gastrointestinal Digestion Upon Solubility and Caco-2 Cell Uptake Assays of Minerals from Infant Formulas

The effect of enzyme amounts used in gastrointestinal in vitro digestion upon the solubility and Caco-2 cell uptake of calcium, iron and zinc from infant formulas (IFs) was studied. Different amounts of enzymes (g enzyme/g IF), pepsin (0.002 and 0.048), pancreatin (0.0005, 0.002 and 0.01) and bile extract (0.003, 0.125 and 0.0625) were assayed. Mineral soluble contents and mineral uptakes by Caco-2 cells were affected by the enzyme amounts used in digestion. Caco-2 cell mineral uptake assays are used to estimate the bioavailability of minerals from foods. The uptake of minerals by Caco-2 cells can be affected by several factors – particularly the conditions of the in vitro digestion process and the growth conditions of the cell culture. Therefore, a standardisation of the assays conditions is required to obtain reproducible results. Resting barley (Hordeum vulgare L.) grains contain acid-proteinase activity. The corresponding enzyme was purified from grain extracts by affinity chromatography on a pepstatin-Sepharose column. The pH optimum of the affinity-purified enzyme was between 3.5 and 3.9 as measured by hemoglobin hydrolysis and the enzymatic activity was completely inhibited by pepstatin a specific inhibitor of aspartic proteinases (EC 3.4.23). International audience; The endoplasmic reticulum (ER) chaperone binding protein (BiP) binds exposed hydrophobic regions of misfolded proteins. Cycles of ATP hydrolysis and nucleotide exchange on the ATPase domain were shown to regulate the function of the ligand-binding domain in vitro. Here we show that ATPase mutants of BiP with defective ATP-hydrolysis (T46G) or ATP-binding (G235D) caused permanent association with a model ligand, but also interfered with the production of secretory, but not cytosolic, proteins in vivo.
