Valorization of natural drug products : from extraction to encapsulation

This thesis was carried out at the Laboratory of Pharmacognosy and the Laboratory of Pharmaceutical technology, at the UFR Sciences de Santé, circonscription Pharmacy, in the University of Burgundy Franche-Comté, to find new natural molecules to encapsulate. First of all, we focused on the natural saponins from plants of the Vietnam biodiversity and the french horticulture, belonging to the three genera Dracaena, Cordyline (Asparagaceae) and Weigela (Caprifoliaceae). Encapsulation of Xenon by a Self-Assembled Fe4L6 Metallosupramolecular Cage

We report (129)Xe NMR experiments showing that a Fe4L6 metallosupramolecular cage can encapsulate xenon in water with a binding constant of 16 M(-1). The observations pave the way for exploiting metallosupramolecular cages as economical means to extract rare gases as well as (129)Xe NMR-based bio-, pH, and temperature sensors. Xe in the Fe4L6 cage has an unusual chemical shift downfield from free Xe in water. The exchange rate between the encapsulated and free Xe was determined to be about 10 Hz, potentially allowing signal amplification via chemical exchange saturation transfer. Killer toxin-secreting double-stranded RNA mycoviruses in the yeasts Hanseniaspora uvarum and Zygosaccharomyces bailii.

Killer toxin-secreting strains of the yeasts Hanseniaspora uvarum and Zygosaccharomyces bailii were shown to contain linear double-stranded RNAs (dsRNAs) that persist within the cytoplasm of the infected host cell as encapsidated virus-like particles. The aim of this work was a better understanding of the effect of the aroma and/or lipid particles incorporation on the structure and physico-chemical properties of film-forming matrices based on iota-carrageenan or sodium alginate. Microstructure, emulsion granulometry, thermal and mechanical properties were assessed as a function of n-hexanal and/or lipid presence. Selection and integration of a new immunomodulatory probiotic strain in a food supplement

Probiotics are live microorganisms that can act on the immune system at specific sites in the gastro-intestinal tract. From a technological point of view, the development of a dietary supplement containing viable and functional probiotics requires the control of the selection and production steps. In a first time, we have developed specific and sensitive screening tests to determine the ability of bacteria to be dried and the origin of its sensitivity. The encapsulation techniques are applied in various fields and for various applications. The layer-by-layer self-assembly (LbL), one of the encapsulation techniques, is built by the electrostatic attraction between oppositely charged polyelectrolytes, and the topmost layer determines essentially the sur-face properties of the edifice. Occurrence of non-target-site-based resistance to ALS inhibitors in the broadleaf weed Papaver rhoeas (corn poppy).

The vast majority of reported cases of resistance to ALS inhibitors in broadleaves is due to the selection of mutant, herbicide-resistant ALS alleles carrying a mutation at one of a few ALS codons that can be easily identified (ALS-based resistance). Non-ALS-based resistance (non-target-site resistance, NTSR), considered to be endowed by differences in the expression of many genes, has essentially been reported in grasses and hardly ever in broadleaves.
