Toward engineering efficient peptidomimetics. Screening conformational landscape of two modified dehydroaminoacids

Effective peptidomimetics should posses structural rigidity and appropriate interaction pattern leading to potential spatial and electronic matching to the target receptor site. Rational design of such small bioactive molecules could push chemical synthesis and molecular modeling toward faster progress in medicinal chemistry. Efficient Synthesis and X-ray Structure of [1,2,4]Triazolo[4,3-a]pyridines via Oxidative Cyclization Using N-Chlorosuccinimide (NCS)

Triazolopyridines are a family of compounds that, owing to their biological activity, have many pharmaceutical applications. In this study, 3-(pyridine-4-yl)-[1,2,4]triazolo[4,3-a]pyridine and 6-bromo-3-(pyridine-4-yl)-[1,2,4]triazolo[4,3-a]pyridine were synthesized by using the chlorinated agent NCS for hydrazones under very mild conditions. The characterization of these compounds was achieved by 1H NMR, 13C NMR, FTIR, MS and X-ray diffraction. The compound 3-(pyridine-4-yl)-[1,2,4]triazolo[4,3-a]pyridine was crystallized in the monoclinic space group P 21/c with a = 15.1413(12), b = 6.9179(4), c = 13.0938(8) Å, β = 105.102(6)°, V = 1324.16(16)Å3, Z = 4, and R = 0.0337. NMR Spectroscopic and quantum chemical characterization of the (E)- and (Z)- isomers of the penta-1,3-dienyl-2-cation

Dilute solutions of the (E )− and (Z )− isomers of pent-1,3-dienyl-2-cations (1) were obtained from reaction of 4-chloro-1,2-pentadiene (2) with SbF5 in SO2ClF/SO2F2 at −135 °C using high-vacuum co-condensation techniques. The experimental NMR spectra of the mixture of the two isomers were compared with quantum chemical 13C NMR chemical shift calculations at HF-SCF, MP2, CCSD and CCSD(T) levels using MP2/tzp geometries. 1H and 13C NMR Measurements of Dimethylchloroformiminium Chloride and Dimethylformamide Hydrochloride

Dual proton and carbon-13 NMR spectra of dimethylchloroformiminium chloride 1 and dimethylformamide hydrochloride 2 in CDCl3 at different temperatures have been measured and the results compared to proton NMR data of dimethyl formamide 3. The energy barriers of internal rotation about C-N bond in 1 and 2 were considerably lower than that of 3. The values of E were 39.3 for 1, 55.9 for 2 and (lit. value) 89.3 for 3 (in kJ/mole).

Abstract Three dithiocarbamatonickel(II) complexes, [Ni(bzbudtc)2] (1), [Ni(bzbudtc)(PPh3)(NCS)] (2) and [Ni(bzbudtc)(PPh3)(CN)] (3) (where, bzbudtc = the N-benzyl-N-butyldithiocarbamato anion and PPh3 = triphenylphosphine), were prepared. All three complexes were analyzed by UV–Vis, IR and NMR (1H, 13C and 31P) spectra. The 13C NMR spectra of complexes 1–3 show the most crucial thioureide signal at around 200 ppm. A significant deshielding observed for the 31P signals in 2 and 3 reveals the effective bonding of phosphorus to the metal center. The 1H and 13C NMR spectral analysis of synthetic (−)-methylthyrsiflorin A and 10 scopadulan precursors is reported.
