Aqueous two-phase system cold-set gelation using natural and recombinant probiotic lactic acid bacteria as a gelling agent

The present study aimed to entrap probiotic lactic acid bacteria (LAB) in a sodium alginate and sodium caseinate aqueous two-phase gel system. The natural acidifying properties of two therapeutic probiotic LAB were exploited to liberate calcium ions progressively from calcium carbonate (CaCO3), which caused the gelation of the co-existing phases. Bi-biopolymeric matrix gelation of GDL/CaCO3 or LAB/CaCO3 was monitored by dynamic rheological measurements, and the final gels were characterized by frequency dependence measurements and confocal laser scanning microscopy. Phosphorylation of the DNA repair protein APE/REF-1 by CKII affects redox regulation of AP-1

The DNA repair protein apurinic endonuclease (APE/Ref-1) exerts several physiological functions such as cleavage of apurinic/apyrimidinic sites and redox regulation of the transcription factor AP-1, whose activation is part of the cellular response to DNA damaging treatments. Here we demonstrate that APE/Ref-1 is phosphorylated by casein kinase II (CKII). This was shown for both the recombinant APE/Ref-1 protein (Km=0.55 mM) and for APE/Ref-1 expressed in COS cells. Phosphorylation of APE/Ref-1 did not alter the repair activity of the enzyme, whereas it stimulated its redox capability towards AP-1, thus promoting DNA binding activity of AP-1. Selectivity analysis of protein kinase CK2 inhibitors DMAT, TBB and resorufin in cisplatin-induced stress responses.

Although many inhibitors are available, exhibiting remarkable potency, the major challenge is their selectivity. Here we show that the protein kinase CK2 inhibitors DMAT, TBB and resorufin differ in their selectivity against PI3K family members, since PI3K and DNA-PK are subject to inhibition by DMAT and TBB, however, not by resorufin. Elongator is a conserved protein complex comprising six different polypeptides that has been ascribed a wide range of functions, but which is now known to be required for modification of uridine residues in the wobble position of a subset of tRNAs in yeast, plants, worms and mammals. In previous work, we showed that Elongator's largest subunit (Elp1; also known as Iki3) was phosphorylated and implicated the yeast casein kinase I Hrr25 in Elongator function. In a previous work (1), we found that a combination of liquid-over-agar-solidified MS (2) modified medium was superior to either all liquid or all solid medium for culture of Digitalis obscura L. mesophyll protoplasts. In order to achieve optimum culture conditions we now report on the influence of a different gelling agent (agarose) as well as the effect of several nutrient media on D. obscura mesophyll protoplasts development.

Isolation of oligopeptides from the water-soluble extract of goat cheese and their identification by mass spectrometry

A procedure for the separation and identification of small peptides from the water-soluble fraction of a goat cheese was developed. The water-soluble extract was ultrafiltered (1000 Da membrane cutoff), and peptides were isolated by sequential chromatography: size exclusion chromatography (HPLC-grade water), anion exchange chromatography (phosphate buffer gradient), and semipreparative reverse-phase high-performance liquid chromatography (water/acetonitrile gradient). Über das Verhalten der Niere nach Injektion größerer Mengen von Casein und substituierten Caseinen

Es wurden die funktionellen und morphologischen Reaktionen der Nieren von Hunden, Kaninchen und Ratten auf Infusionen von Losungen verschieden dargestellter und substituierter Caseine untersucht. Funktionelle Veranderungen konnten nur beim formylsubstituierten Casein beobachtet werden. Die beobachteten morphologischen Veranderungen wurden in ihrem zeitlichen Ablauf verfolgt und kurvenmasig dargestellt. Die einzelnen Praparate unterscheiden sich dabei deutlich. Unsere Befunde sprechen fur eine Fahigkeit der Tubulusepithelien, glomerular abgeschiedene Proteine durch Resorption eliminieren zu konnen.

Does the addition of caseinophosphopeptides or milk improve zinc in vitro bioavailability in fruit beverages?

Abstract The influence of caseinophosphopeptides (CPPs) added to fruit beverage versus milk based fruit beverage upon zinc retention, transport, and uptake, as well as the influence of Fe supplementation, were studied using a combined simulated gastrointestinal digestion/Caco-2 cell system. Zinc retention, transport, and uptake of milk based fruit beverage was 4- to 5-fold greater than that of fruit beverages with or without CPPs – no statistically significant differences being observed in relation to the presence or absence of CPPs.
