Metallothionein Gene Family in the Sea Urchin Paracentrotus lividus: Gene Structure, Differential Expression and Phylogenetic Analysis

Metallothioneins (MT) are small and cysteine-rich proteins that bind metal ions such as zinc, copper, cadmium, and nickel. In order to shed some light on MT gene structure and evolution, we cloned seven Paracentrotus lividus MT genes, comparing them to Echinodermata and Chordata genes. Moreover, we performed a phylogenetic analysis of 32 MTs from different classes of echinoderms and 13 MTs from the most ancient chordates, highlighting the relationships between them. Since MTs have multiple roles in the cells, we performed RT-qPCR and in situ hybridization experiments to understand better MT functions in sea urchin embryos. Traditionally, the family Campulidae has been associated either with the family Fasciolidae, parasites of ruminants, or the Acanthocolpidae, parasites of fishes, based on morphological similarities. Since morphology does not seem to resolve clearly the problem of the relationships of campulids, we have used the sequences of the 18S rRNA gene of the campulids Zalophotrema hepaticum, Campula oblonga and Nasitrema globicephalae, the fasciolid Fasciola hepatica, the acanthocolpid Stephanostomum baccatum and the outgroup Schistosoma mansoni to infer a phylogeny. In order to study the defense strategies activated by Paracentrotus lividus embryos in response to sub-lethal doses of CdCl2, we compared the induced transcripts to that of control embryos by suppression subtractive hybridization technique. We isolated five metallothionein (MT) cDNAs and other genes related to detoxification, to signaling pathway components, to oxidative, reductive and conjugative biotransformation, to RNA maturation and protein synthesis. The oldest echinoderms and first cornute stylophorans ever reported from Korea are described, based on more than 40 specimens collected from the Late Cambrian of the Taebaeksan Basin. New material doubles the number of stylophorans described from Asia and the number of specimens of Late Cambrian stylophorans recorded throughout the world. Three different cornutes are identified: Sokkaejaecystis serrata n. gen. and sp. and two genus and species indeterminate forms A and B. En la presente Tesis Doctoral se analizan diferentes parámetros implicados en las relaciones parásito-hospedador en helmintiasis intestinales mediante la utilización del modelo experimental Echinostoma caproni-roedor. Con esta finalidad se estudian diversos aspectos inmunológicos, además de las alteraciones intestinales inducidas por la infección en dos sistemas parásito-hospedador: E. caproni-ratón y E. caproni-rata. Los resultados obtenidos ponen de manifiesto que el IFN-γ y el óxido nítrico son mediadores fundamentales en el desarrollo de infecciones crónicas en ratón, induciendo disfunción mitocondrial en las células del epitelio intestinal e hiperplasia tisular. The essay explores the reception of Rossini’s Italian operas in the writings of Étienne-Jean Delécluze. A leading figure in the Parisian cultural milieu of his time, Delécluze reviewed performances at the Théâtre Italien for the influential «Journal des débats» between 1832 and 1863. He developed an interpretative frame based on the idea that the Théâtre-Italien should function as as a “school of singing” for Paris, and identified Rossini’s operas as canonical texts that such a school should preserve and promote, in opposition to more recent trends originating from Italy. The impact of these processes on marine organisms will depend on their ability to cope with those changes, particularly the maintenance of calcium carbonate structures.
