Abstract This paper describes the on-line sample pretreatment and analysis of proteins and peptides with a poly(methylmethacrylate) (PMMA) microfluidic device (IonChip™). This chip consists of two hyphenated electrophoresis channels with integrated conductivity detectors. The first channel can be used for sample preconcentration and sample clean-up, while in the second channel the selected compounds are separated. Isotachophoresis (ITP) combined with zone electrophoresis (CZE) was used to preconcentrate a myoglobin sample by a factor of about 65 before injection into the second dimension and to desalt a mixture of six proteins with 100 mM NaCl. Here we show the fabrication of the Luminometric Sub-nanoliter Droplet-to-droplet Array (LUMDA chip) by inkjet printing. The chip is easy to be implemented and allows for a multiplexed multi-step biochemical assay in sub-nanoliter liquid spots. This concept is here applied to the integral membrane enzyme CYP3A4, i.e. the most relevant enzymatic target for phase I drug metabolism, and to some structurally-related inhibitors.
Genome-wide detection of copy-number variations in local cattle breeds

The aim of the present study was to identify copy-number variations (CNVs) in Cinisara (CIN) and Modicana (MOD) cattle breeds on the basis of signal intensity (logR ratio) and B allele frequency of each marker, using Illumina’s BovineSNP50K Genotyping BeadChip. The CNVs were detected with the PennCNV and SVS 8.7.0 software and were aggregated into CNV regions (CNVRs). PennCNV identified 487 CNVs in CIN that aggregated into 86 CNVRs, and 424 CNVs in MOD that aggregated into 81 CNVRs. SVS identified a total of 207 CNVs in CIN that aggregated into 39 CNVRs, and 181 CNVs in MOD that aggregated into 41 CNVRs. Salinity affects nitrate removal and microbial composition of denitrifying woodchip bioreactors treating recirculating aquaculture system effluents

Abstract This study investigated the effect of salinity on microbial composition and denitrification capacity of woodchip bioreactors treating recirculating aquaculture system (RAS) effluents. Twelve laboratory-scale woodchip bioreactors were run in triplicates at 0, 15, 25, and 35 ppt salinities, and water chemistry was monitored every third day during the first 39 days of operation. Microbial communities of the woodchips bioreactors were analyzed at the start, after one week, and at the end of the trial. Woodchip bioreactors removed nitrate at all salinities tested. Continental shelf island systems, created by rising sea levels, provide a premier setting for studying the effects of past fragmentation, dispersal, and genetic drift on taxon diversification. We used phylogeographical (nested clade) and population genetic analyses to elucidate the relative roles of these processes in the evolutionary history of the Aegean Nigella arvensis alliance (= ‘coenospecies’). Abstract An overview of the population histories of three insular realms (Gargano palaeo-archipelago, Sardinia–Maritime Tuscany palaeo-bioprovince and the Sicilian insular complex) during the Late Miocene and Quaternary are here presented. The complexity of biodiversity changes in the islands is analysed to propose an interpretation of origination and extinction patterns. The study highlighted several important aspects of insular faunas. Evolutionary radiations were found to contribute significantly only to the Gargano faunal diversity, likely because the area was an archipelago at the time. The Pico do Fogo volcano, in the Cape Verde Archipelago off the western coasts of Africa, has been the most active volcano in the Macaronesia region in the Central Atlantic, with at least 27 eruptions during the last 500 years.
