A novel microtubule de-stabilizing complementarity-determining region C36L1 peptide displays antitumor activity against melanoma in vitro and in vivo

AbstractShort peptide sequences from complementarity-determining regions (CDRs) of different immunoglobulins may exert anti-infective, immunomodulatory and antitumor activities regardless of the specificity of the original monoclonal antibody (mAb). In this sense, they resemble early molecules of innate immunity. C36L1 was identified as a bioactive light-chain CDR1 peptide by screening 19 conserved CDR sequences targeting murine B16F10-Nex2 melanoma. The 17-amino acid peptide is readily taken up by melanoma cells and acts on microtubules causing depolymerization, stress of the endoplasmic reticulum and intrinsic apoptosis. Synthesis and biological evaluation of novel peptidomimetics as rhodesain inhibitors

Novel rhodesain inhibitors were developed by combining an enantiomerically pure 3-bromoisoxazoline warhead with a 1,4-benzodiazepine scaffold as specific recognition moiety. All compounds were proven to inhibit rhodesain with Ki values in the low-micromolar range. Their activity towards rhodesain was found to be coupled to an in vitro antitrypanosomal activity, with IC50 values ranging from the mid-micromolar to a low-micromolar value for the most active rhodesain inhibitor (R,S,S)-3. All compounds showed a good selectivity against the target enzyme since all of them were proven to be poor inhibitors of human cathepsin L. Detection of natural killer T cells in mice infected with Rickettsia conorii.

Little information is available regarding the role of natural killer T (NKT) cells during the early stage of Rickettsia conorii infection. Herein, C3H/HeN mice were infected with the Malish 7 strain of R. conorii. Splenocytes from these mice were analysed in the early stage of the infection by flow cytometry and compared with uninfected controls. Our results showed an increase in NKT cells in infected mice. Additionally, NKT interleukin (IL)-17(+) cells increased three days after infection, together with a concurrent decrease in the relative amount of NKT interferon (IFN)-γ(+) cells. The Effect of Mechanocatalytic Pretreatment on the Structure and Depolymerization of Willow

In this study, the effect of a mechanocatalytic pretreatment on the structure of willow and sugar release from pretreated willow was explored. In the mechanocatalytic approach, the pretreatment consists of solvent-free impregnation with sulfuric acid and a mechanical treatment with ball milling. Willow sawdust and pretreated samples were analyzed with field emission scanning electron microscope and X-ray diffraction. The products in the sugar solution were determined as the total reducing sugars with the 3,5-dinitrosalicylic acid method and monosaccharides with capillary electrophoresis. Una statua recentemente ricomposta nell'Antiquarium di Solunto, ad un esame dettagliato, risulta essere un assemblaggio di parti pertinenti in origine ad almeno due statue improntate al cd. tipo della Kore di Prassitele, una delle quali pare avere inoltre subito la sostituzione del ritratto originario con una nuova effigie femminile. Lo studio iconografico-stilistico consente di riconoscere nel ritratto attualmente preservato, in piccola parte, sulla fronte Agrippina Maggiore e nel frammento posteriore un'effigie ugualmente di età neroniana. The material dimension of cult practices and ritual communication in ancient sanctuaries is the central issue of all contributions gathered in the book, which has been framed within three complementary areas of enquiry. Firstly, the architectural settings of sanctuaries was examined beyond temple buildings to appraise the wider context of their structural and spatial complexity. Secondly, the material culture of votive deposition and religious feasting is analysed in terms of performative characteristics and through the lens of anthropological approaches. Thirdly, the significance of gender in cultic practice is investigated in light of the fresh data retrieved from the field.

Selection of spawning substratum by European river lampreys (Lampetra fluviatilis) in experimental tanks

The selection of spawning substratum by the river lamprey (Lampetra fluviatilis) was studied in two experimental tanks with different flow conditions. In both tanks, four gravel sizes mixed with 15% sand were available to the test animals. In the tank with the lower current speed, lampreys selected in favour of the finest (2–8 mm) gravel size available against gravel sizes 4–20 mm and 8–40 mm.
