Oxidative stress-induced glucocorticoid resistance is prevented by dual PDE3/PDE4 inhibition in human alveolar macrophages

2011

Summary Background Oxidative stress is present in airway diseases such as severe asthma or Chronic Obstructive Pulmonary Disease and contributes to the low response to glucocorticoids through the down-regulation of histone deacetylase (HDAC) activity. Objective To study the effects of the phosphodiesterase (PDE)-3 and 4 inhibitors and their combination vs. glucocorticoids in a model of lipopolysaccharide (LPS)-induced cytokine release in alveolar macrophages under oxidative stress conditions. Methods Differentiated U937 or human alveolar macrophages were stimulated with H2O2 (10–1000 μm) or cigarette smoke extract (CSE, 0–15%) for 4 h before LPS (0.5 μg/mL, 24 h) addition. In other experime…

Corneal opacity and copper levels of the Lewis syndrome after systemic chemotherapy

2020

Abstract Purpose To report a female patient of biclonal Lewis syndrome which consists of a trias: biclonal gammopathy of undetermined significance, paraproteinemic keratopathy in form of a brownish discoid opacification at the level of Descemet's membrane and hypercupremia. After several years there was a conversion to multiple myeloma. Systemic chemotherapy led to a complete remission of multiple myeloma and to a normalization of the copper level in the blood that lasted five years. The corneal opacification remained unchanged. Observations A currently 66-year-old woman suffered from biclonal Lewis syndrome. On both eyes there is a central discoid yellow-brownish discoloration in the Pre-D…

Molecular and Translational Classifications of DAMPs in Immunogenic Cell Death

2015

The immunogenicity of malignant cells has recently been acknowledged as a critical determinant of efficacy in cancer therapy. Thus, besides developing direct immunostimulatory regimens, including dendritic cell-based vaccines, checkpoint-blocking therapies, and adoptive T-cell transfer, researchers have started to focus on the overall immunobiology of neoplastic cells. It is now clear that cancer cells can succumb to some anticancer therapies by undergoing a peculiar form of cell death that is characterized by an increased immunogenic potential, owing to the emission of the so-called "damage-associated molecular patterns" (DAMPs). The emission of DAMPs and other immunostimulatory factors by…

Synthesis of biotin-labelled dexamethasone derivatives. Novel hormone-affinity probes.

1983

A new, general methodology for 'sandwich' affinity chromatography of steroid hormone receptors is proposed, the part purification of the human spleen tumor glucocorticoid receptor is quoted as an illustration. 9-Fluoro-16 alpha-methyl-11 beta, 17-dihydroxy-1,4-androstadiene-3-one-17 beta-carboxylic acid was coupled to biotin using pentamethylenediamine (BioDex 1) as a spacer. The bifunctional derivative binds to glucocorticoid receptors and avidin-Sepharose and efficiently protects the glucocorticoid receptor against inactivation when previously added during homogenisation. We have standardized the capacity and optimum conditions for elution of receptor-BioDex-1 complexes which are bound to…

Phosphorus-Chalcogen Ring Expansion and Metal Coordination

2017

The reactivity of 4-membered (RPCh)2 rings (Ch = S, Se) that contain phosphorus in the +3 oxidation state is reported. These compounds undergo ring expansion to (RPCh)3 with the addition of a Lewis base. The 6-membered rings were found to be more stable than the 4-membered precursors, and the mechanism of their formation was investigated experimentally and by density functional theory calculations. The computational work identified two plausible mechanisms involving a phosphinidene chalcogenide intermediate, either as a free species or stabilized by a suitable base. Both the 4- and 6-membered rings were found to react with coinage metals, giving the same products: (RPCh)3 rings bound to the…

Ripostigli “misti” dalla Sicilia centro-meridionale

2011

It is remarkable the discovery in the hinterland of Sicily of three “mixed” hoards, including not only coins, but also of bronze scraps and small fragmentary objects. They demonstrate that in these centers, still in the 4th century BC, the coin was considered a worthless piece of metal and that other bronze objects were used as medium of exchange. The subject of further study is the so-called closet of Milocca-Milena (IGCH 2162), which is attributed to the site of Monte Raffe

Statistical Modelling and Risk Analysis of Bitcoins Exchange Rate

2018

Master's thesis Business Administration BE501 - University of Agder 2018 Bitcoin is a phenomenon that is new and there is little information on how and why it behaves as volatile as it does. This thesis uses existing data on Bitcoin’s exchange rate to estimate a model that describes the pattern and use it in a financial risk analysis. We also aim to contribute as a foundation for further studies in this field. The statistical properties of the log-return of the exchange rate are analysed and it is deemed to be iid. From the eleven distributional candidates we study is the fitted skew generalised t distribution proven to represent the data best after evaluation by criteria and statistics. Th…

Glucocorticoid-sensitive hereditary inclusion body myositis.

1996

We report a hereditary muscle disorder with features of inclusion body myositis (IBM) in two adult sisters with slowly progressive asymmetrical muscle weakness. The findings of light microscopic and ultrastructural investigations of muscle biopsy specimens were consistent with a diagnosis of IBM. Both patients improved and stabilized on immunosuppressive treatment with corticosteroids and azathioprine. This differentiates our patients from other sporadic and familial cases of IBM. Clinical and histological features are described and compared with those of other previously reported families with IBM.

Viewpoint: On the hysteresis in the human Achilles tendon.

2012

This viewpoint was stimulated by two observations: 1) the statistical skewness whereby numerous articles have reported tendon stiffness and Young9s modulus, but far fewer have reported tendon hysteresis; 2) in vivo human studies seem very often to report hysteresis values greater than 10%, suggesting either that there are methodological differences between human and animal studies, or that human tendons have a much poorer capacity to store and reutilize elastic energy. In this article we focus on the healthy human Achilles/gastrocnemius tendon (AT) since it has an important locomotor function and clearly a low AT hysteresis would allow elastic recoil for efficient locomotion. We discuss tha…
