Immunotherapy targeting colon cancer stem cells

2010

In the last 10 years, cancer stem cells have interested the scientific community because this small tumorigenic population is also associated with tumor progression in human patients and specific targeting of cancer stem cells could be a strategy to eradicate cancers currently resistant to conventional therapy. Clinical studies have recently demonstrated that adding immune therapy to chemotherapy has survival benefits in comparison with chemotherapy alone that can sensitize tumors to immune cell-mediated killing (e.g., increasing sensitivity of tumor cells to subsequent cytotoxicity by T cells via upregulation of death receptors DR5 and Fas). However, loss of MHC molecules is often observe…

Anti-Human CD9 Fab Fragment Antibody Blocks the Extracellular Vesicle-Mediated Increase in Malignancy of Colon Cancer Cells.

2022

Intercellular communication between cancer cells themselves or with healthy cells in the tumor microenvironment and/or pre-metastatic sites plays an important role in cancer progression and metastasis. In addition to ligand–receptor signaling complexes, extracellular vesicles (EVs) are emerging as novel mediators of intercellular communication both in tissue homeostasis and in diseases such as cancer. EV-mediated transfer of molecular activities impacting morphological features and cell motility from highly metastatic SW620 cells to non-metastatic SW480 cells is a good in vitro example to illustrate the increased malignancy of colorectal cancer leading to its transformation and aggressive b…

REAL HUMANS? : Affective imaginaries of the human and its Others in the Swedish TV series Äkta människor

2019

According to the Swedish science fiction TV series Äkta människor (Real Humans, SVT and Matador film 2012-2014), humanoid robots called “hubots” are replacing the human workforce in care work and assembly line industries. Against the backdrop of current debates about immigration and citizenship in the Nordic countries, this article does a close, contextual reading of the series, exploring how the hubots influence work and family life. We are particularly interested in how hubots tie in with the cultural circulation of affect in relation to Otherness and how responses towards the “not-quite” human or dehumanized Other are negotiated in the present-day Nordic cultural imaginaries. What kinds …

Cetuximab +/- chemotherapy enhances dendritic cell-mediated phagocytosis of colon cancer cells and ignites a highly efficient colon cancer antigen-sp…

2012

Cetuximab is a human/mouse chimeric IgG1 monoclonal antibody (mAb) to epidermal growth factor receptor, approved for colorectal carcinoma treatment in combination with chemotherapy. The immune-mediated effects elicited by its human fraction of crystallization moiety might critically contribute to the overall anti-tumor effectiveness of the antibody. We therefore investigated cetuximab ability to promote colon cancer cell opsonization and phagocytosis by human dendritic cells (DCs) that are subsequently engaged in antigen-cross presentation to cytotoxic T-lymphocyte (CTL) precursors. Human colon cancer cell lines were evaluated for susceptibility to DC-mediated phagocytosis before and after …

Acylated Triterpene Saponins from Atroxima libericaStapf

2011

The four new acylated triterpene saponins 1–4, isolated as two pairs of isomers and named libericosides A1/A2 and B1/B2, one pair of isomers 5/6, the (Z)-isomer libericoside C2 (5) being new, one new sucrose ester, atroximoside (7), and eight known compounds were isolated from the roots of Atroxima liberica by repeated MPLC and VLC on normal and reversed-phase silica gel. Their structures were elucidated on the basis of extensive 1D- and 2D-NMR studies (1H- and 13C-NMR, DEPT, COSY, TOCSY, NOESY, HSQC, and HMBC) and mass spectrometry as 3-O-β-D-glucopyranosylpresenegenin 28-{O-α-L-arabinopyranosyl-(13)-O-β-D-xylopyranosyl-(14)-O-α-L-rhamnopyranosyl-(12)-4-O-[(E)-3,4-dimethoxycinnamoyl]-β-D-f…

Phytochemical analysis of two Weigela florida cultivars, “Pink Poppet” and “Jean’s Gold”

2020

Abstract Nine different oleanane-type glycosides were extracted and isolated by various chromatographic methods from two Weigela florida cultivars, “Pink Poppet” and “Jean’s Gold”. From the roots of W. florida “Pink Poppet”, three monodesmosidic oleanolic acid saponins 1, 4, 5 were obtained, together with one hederagenin ester 6 from the leaves, and six bidesmosidic saponins 2, 3, 6-9 were isolated from the leaves of W. florida “Jean’s Gold”. Among compounds 1-9, three were previously undescribed (1-3) and six (4-9) were already published in the literature. Their structures were assigned by spectroscopic analysis mainly 2D NMR and mass spectrometry (ESI-MS). The cytotoxicity of the isolated…
