Isolated lung perfusion with chemotherapy in a pig model

2014

Introduction: The isolated lung perfusion (ILP) is an experimental technique which main objective is to deliver high dose of cytotoxic agent to the lung tissue without systemic exposure. The thesis took place in three stages: first stage, setting in vitro the chemotherapy the most efficient against colorectal cancer (CCR) cells in 30 min. Second stage, develop the ILP technique in a pig model. Third stage, lead a dose escalation study with chemotherapy by ILP.Methods: First stage, efficacy of various cytotoxic molecules against a panel of human CCR cell lines was tested in vitro after a 30-minute exposure. Second stage, pigs were treated with chemotherapy delivered by ILP during 30 minutes …

ChemInform Abstract: Cyclooxygenase Inhibitors - Current Status and Future Prospects

2010

Prostaglandins are formed from arachidonic acid by the action of cyclooxygenase and subsequent downstream synthetases. Two closely related forms of the cyclooxygenase have been identified which are now known as COX-1 and COX-2. Both isoenzymes transform arachidonic acid to prostaglandins, but differ in their distribution and their physiological roles. Meanwhile, the responsible genes and their regulation have been clarified. COX-1, the pre-dominantly constitutive form of the enzyme, is expressed throughout the body and performs a number of homeostatic functions such as maintaining normal gastric mucosa and influencing renal blood flow and platelet aggregation. In contrast, the inducible for…

Prognostic value of neurotrophic growth factor (BDNF) in patients with colorectal cancer during chemotherapy

2019

INTRODUCTION: The brain-derived neurotrophic factor (BDNF) is a protein belonging to neurotrophins that plays a key role in the proper development and functioning of the mammalian central nervous system. Previous studies have focused on assessment of the BDNF concentration in blood serum as a potential biomarker in neurological disorders. Recently, the BDNF signalling pathway has been recognised as a potential target for anticancer drugs, while its receptor (TrkB) as an oncogene in colorectal cancer cells. Despite the significant role in carcinogenesis, there are few studies on BDNF as a biomarker in colorectal cancer. MATERIAL AND METHODS: The study included 25 patients with clinically and…

Simultaneous Color Contrast in Goldfish— a Quantitative Study

1997

AbstractA set of 9–15 colored test fields was presented to goldfish. In Experiment 1, test field hues ranged from green through yellow to red; in Experiment 2, the hues varied from blue through gray to yellow. In the training conditions, the test fields were presented with a gray or black surround. The fish learned to choose one intermediate test field hue by rewarding them with food. In the test conditions, the color of the surround was changed from gray to green, or red (Experiment 1), and from black to blue, or yellow (Experiment 2). The choice behavior of the goldfish changed substantially: one of the test fields other than the training test field was preferred. Direction and strength o…

<title>Stereoacuity determination at changing contrast of colored stereostimuli</title>

2003

Studies are focused on design and appraisal of an objective test for assessment of the stereovision quality in unfavorable conditions. Stereostimuli of different colors are used while the contrast of one of the stimulus being varied. Tests are based on principles of black-and-white and two primary color random dot stereotests. Experiments are divided by the method of stimuli display and separation: 1) stereoeffect is obtained haploscopically - by use of spectacles with color filters (blue and red) or prisms, 2) stimuli separation is obtained by liquid crystal shutters when both eye stimuli are demonstrated with a different delay. The stereovision threshold is determiend at different stimuli…

A perspective analysis: microRNAs, glucose metabolism, and drug resistance in colon cancer stem cells

2021

Metabolism sustains the stemness of Cancer Stem Cells (CSCs), affecting, in turn, tumor heterogeneity, metastatic potential, and therapy resistance. Therefore, it is appealing to target CSCs metabolism as a new therapeutic approach. Consequently, we paid considerable attention to the anti-apoptotic microRNA miR-483-3p, that we reported being regulated by glucose metabolism in liver cancer cells. We investigated the therapeutic potential of targeting miR-483-3p by using the anti-glucose metabolism 2-deoxyglucose (2-DG) molecule in tumor Xenograft mouse model originating from two different Colon-Cancer Stem Cell lines (CCSC lines). We show that 2-DG treatment does not affect CCSCs during tumo…

Adipose stromal cells promote the transition of colorectal cancer cells toward a mesenchymal-like phenotype

2021

ABSTRACT Colon cancer progression is among the risks that increase with obesity. We have recently unveiled the molecular mechanism by which adipose tissue-released molecules, HGF and IL-6, make colorectal cancer (CRC) cells acquiring mesenchymal traits. Targeting of adipose-derived factors abrogate the metastatic potential of CRC stem cells (CR-CSCs) in obese patients.

Dual Inhibition of Myc Transcription and PI3K Activity Effectively Targets Colorectal Cancer Stem Cells

2022

Despite advances in the curative approach, the survival rate of advanced colorectal cancer (CRC) patients is still poor, which is likely due to the emergence of cancer cell clones resistant to the available therapeutic options. We have already shown that CD44v6-positive CRC stem cells (CR-CSCs) are refractory toward standard anti-tumor therapeutic agents due to the activation of the PI3K pathway together with high HER2 expression levels. Tumor microenvironmental cytokines confer resistance to CR-CSCs against HER2/PI3K targeting by enhancing activation of the MAPK pathway. Here, we show that the CSC compartment, spared by BRAF inhibitor-based targeted therapy, is associated with increased ex…

Colorectal Cancer Stem Cells and Cell Death

2011

Nowadays it is reported that, similarly to other solid tumors, colorectal cancer is sustained by a rare subset of cancer stem-like cells (CSCs), which survive conventional anticancer treatments, hanks to efficient mechanisms allowing escape from apoptosis, triggering tumor recurrence. To improve patient outcomes, conventional anticancer therapies have to be replaced with specific approaches targeting CSCs. In this review we provide strong support that BMP4 is an innovative therapeutic approach to prevent colon cancer growth increasing differentiation markers expression and apoptosis. Recent data suggest that in colorectal CSCs, protection from apoptosis is achieved by interleukin-4 (IL-4) a…
