The dark side of the moon: The PI3K/PTEN/AKT pathway in colorectal carcinoma

2009

Wild-type KRAS status is required but not sufficient to confer sensitivity to anti-epidermal growth factor receptor (EGFR) monoclonal antibodies (mAbs) in colorectal cancer patients. As a consequence, one of the major challenges is to identify, in non-mutant KRAS patients, other markers that can predict lack of response to this therapy. Small series have investigated the clinical effect of PIK3CA mutations on resistance to anti-EGFR mAbs and discrepant results have been observed. Furthermore, PTEN loss in metastases may be predictive of resistance to anti-EGFR mAbs, even if PTEN determination is far from an immediate clinical application. The introduction of modulators of the PI3K/AKT/mTOR …

Palermo bianca

2019

White Palermo may seem like an invisible city; and, however, despite the stratifications and transformations, it is to be counted among the authentic white cities, especially for being one of the capitals of the Mediterranean. White is traceable in some stages of the city's history, confirming the ambiguous origins and mul…

Search for massive colored scalars in four-jet final states in √s = 7 TeV proton–proton collisions with the ATLAS detector

2011

A search for pair-produced scalar particles decaying to a four-jet final state is presented. The analysis is performed using an integrated luminosity of 34 pb[superscript −1] recorded by the ATLAS detector in 2010. No deviation from the Standard Model is observed. For a scalar mass of 100 GeV (190 GeV) the limit on the scalar gluon pair production cross section at 95% confidence level is 1 nb (0.28 nb). When these results are interpreted as mass limits, scalar-gluons (hyperpions) with masses of 100 to 185 GeV (100 to 155 GeV) are excluded at 95% confidence level with the exception of a mass window of width about 5 GeV (15 GeV) around 140 GeV.

Confocal laser endomicroscopy is a new imaging modality for recognition of intramucosal bacteria in inflammatory bowel disease in vivo.

2011

International audience; BACKGROUND AND OBJECTIVES: Interaction of bacteria with the immune system within the intestinal mucosa plays a key role in the pathogenesis of inflammatory bowel disease (IBD). The aim of the current study was to develop a fluorescein-aided confocal laser endomicroscopy (CLE) method to visualise intramucosal enteric bacteria in vivo and to determine the involved mucosal area in the colon and ileum in patients with ulcerative colitis (UC) and Crohn's disease (CD). METHODS: Initially, E coli strains expressing enhanced green fluorescent protein (pEGFP) were endomicroscopically imaged in mice. In addition, ex vivo and in vivo imaging of fluorescent human enteric bacteri…

Endoscopic surveillance in ulcerative colitis: smart biopsies do it better.

2007

Patients with ulcerative colitis (UC) have a significantly higher risk for the development of colitisassociated colorectal cancer. Endoscopic surveillance is recommended. The growth pattern of dysplastic tissue is often multifocal and flat. Thus, significant lesions might be overlooked. Multiple random biopsies are recommended, but cannot eliminate the possibility of missed cancers with severe consequences for the patient. Chromoendoscopy with methylene blue or indigo carmine can unmask flat lesions and may solve this problem. In addition, confocal laser endomicroscopy enables in vivo histology with subcellular resolution during ongoing colonoscopy and leads to highly selected mucosal biops…

Microsatellite Instability in Colorectal Cancer: Time to Stop Hiding!

2011

Colorectal cancer (CRC) is the second cause of cancer-related death worldwide. Surgery constitutes the primary therapy for these tumors, together with chemotherapy that is usually recommended in patients with metastatic primary CRC. Although molecularly distinct entities arising from different physiopathogenic mechanisms - microsatellite (MSI) and chromosomal instability (also called microsatellite stable, MSS) - have been characterized in CRC, there is still no specific therapeutic approach that takes into account disease’s molecular heterogeneity [1]. MSI is observed in 1015% of sporadic CRCs. MSI CRCs displayed particular morphologic features, with greater predilection for the right colo…

Hepatocellular carcinoma and synchronous liver metastases from colorectal cancer in cirrhosis: A case report.

2013

A 68-year-old Caucasian man with hepatitis C virus-related cirrhosis was admitted to our Unit in February 2010 for a diagnostic evaluation of three centimetric hypoechoic focal liver lesions detected by regular surveillance ultrasound. The subsequent computer tomography (CT) led to a diagnosis of unifocal hepatocellular carcinoma (HCC) in VI hepatic segment, defined the other two nodules in the VI and VII segment as suspected metastases, and showed a luminal narrowing with marked segmental circumferential thickening of the hepatic flexure of the colon. Colonoscopy detected an ulcerated, bleeding and stricturing lesion at the hepatic flexure, which was subsequently defined as adenocarcinoma …

Assessment of Tumor Development and Wound Healing Using Endoscopic Techniques in Mice

2010

Mouse models of intestinal inflammation and colon cancer are valuable tools to gain insights into the pathogenesis of the corresponding human diseases. Recently, in vivo mouse endoscopy has been developed, allowing not only the high-resolution monitoring and scoring of experimental disease development, but also enables the investigator to perform manipulations, including local injection of reagents or the taking of biopsies for molecular and histopathologic analyses. Chromoendoscopic staining with methylene blue enables visualization of the crypt structure and allows discrimination between inflammatory and neoplastic changes. The development of endoscopic techniques in live mice opened new …

Metastatic seeding of colon adenocarcinoma manifesting as synchronous breast and chest wall localization: report of a case.

2011

Colon carcinoma rarely metastasizes to the breast and it is usually associated with a poor prognosis. Even rarer is metastatic seeding of colon cancer cells in an intramammary location after surgery. Including a primary breast malignancy in the differential diagnosis of such cases is mandatory. We report a rare case of double seeding implantation of colon adenocarcinoma inside the breast parenchyma and intercostal muscles 6 years after resection of a pulmonary metastasis from colon adenocarcinoma. The metastasis was revealed by the presence of bone metaplasia in the intercostal muscles. We discuss how negative immunostaining for estrogen receptors, progesterone receptors, and HER-2, along w…
