Assembly of Polycomb complexes and silencing mechanisms

Polycomb complexes assemble at their target sites and silence neighboring genes when these are not actively transcribed. The action of these complexes and of Trithorax complexes bound to the Polycomb Response Element establish alternative silent or derepressed states that are remembered through cell division and maintained for the rest of development. Recent results that may help explain the properties of these states are reviewed.

The COVID-19 pandemic is causing a dramatic loss of lives worldwide, challenging the sustainability of our health care systems, threatening economic meltdown, and putting pressure on the mental health of individuals (due to social distancing and lock-down measures). A spontaneous mitonuclear epistasis converging on Rieske Fe-S protein exacerbates complex III deficiency in mice

We previously observed an unexpected fivefold (35 vs. 200 days) difference in the survival of respiratory chain complex III (CIII) deficient Bcs1lp.S78G mice between two congenic backgrounds. Here, we identify a spontaneous homoplasmic mtDNA variant (m.G14904A, mt-Cybp.D254N), affecting the CIII subunit cytochrome b (MT-CYB), in the background with short survival. We utilize maternal inheritance of mtDNA to confirm this as the causative variant and show that it further decreases the low CIII activity in Bcs1lp.S78G tissues to below survival threshold by 35 days of age. FO^2 with one transitive relation is decidable

We show that the satisfiability problem for the two-variable first-order logic, FO^2, over transitive structures when only one relation is required to be transitive, is decidable. The result is optimal, as FO^2 over structures with two transitive relations, or with one transitive and one equivalence relation, are known to be undecidable, so in fact, our result completes the classification of FO^2-logics over transitive structures with respect to decidability. We show that the satisfiability problem is in 2-NExpTime. Decidability of the finite satisfiability problem remains open.

New results for finding common neighborhoods in massive graphs in the data stream model

AbstractWe consider the problem of finding pairs of vertices that share large common neighborhoods in massive graphs. We give lower bounds for randomized, two-sided error algorithms that solve this problem in the data-stream model of computation. Our results correct and improve those of Buchsbaum, Giancarlo, and Westbrook [On finding common neighborhoods in massive graphs, Theoretical Computer Science, 299 (1–3) 707–718 (2004)]

CD95 death-inducing signaling complex formation and internalization occur in lipid rafts of type I and type II cells

We investigated the membrane localization of CD95 in type I and type II cells, which differ in their ability to recruit and activate caspase-8. We found that CD95 was preferentially located in lipid rafts of type I cells, while it was present both in raft and non-raft plasma membrane sub-domains of type II cells. After stimulation, CD95 located in phospholipid-rich plasma membrane was recruited to lipid rafts in both types of cells. Similarly, CD95 cross-linking resulted in caspase-independent translocation of FADD/MORT1 and caspase-8 to the lipid rafts, which was prevented by a death domain-defective receptor. Verbal ordinal classification with multicriteria decision aiding

Abstract Professionals in neuropsychology usually perform diagnoses of patients’ behaviour in a verbal rather than in a numerical form. This fact generates interest in decision support systems that process verbal data. It also motivates us to develop methods for the classification of such data. In this paper, we describe ways of aiding classification of a discrete set of objects, evaluated on set of criteria that may have verbal estimations, into ordered decision classes.
