Novel imine antioxidants at low nanomolar concentrations protect dopaminergic cells from oxidative neurotoxicity.

Strong evidence indicates that oxidative stress may be causally involved in the pathogenesis of Parkinson's disease. We have employed human dopaminergic neuroblastoma cells and rat primary mesencephalic neurons to assess the protective potential of three novel bisarylimine antioxidants on dopaminergic cell death induced by complex I inhibition or glutathione depletion. We have found that exceptionally low concentrations (EC(50) values approximately 20 nM) of these compounds (iminostilbene, phenothiazine, and phenoxazine) exhibited strong protective effects against the toxicities of MPP(+), rotenone, and l-buthionine sulfoximine. This work reports on the valorisation of persimmon (Diospyros kaki Thunb.) for the development of food-grade antiviral coatings against major viral foodborne pathogens, human noroviruses (NoVs) and hepatitis A virus (HAV). Initially, the antiviral activity of polyphenol-rich pectin extracts with abundant non-covalent interactions (PPN), pectin extracts enriched with intact pectin-polyphenol ester and O-glycosyl bonds (PPC) and hydro-ethanolic polyphenol-rich extracts (EPE) was compared. Haptoglobin interacts with apolipoprotein E and beta-amyloid and influences their crosstalk.

Beta-amyloid accumulation in brain is a driving force for Alzheimer's disease pathogenesis. Apolipoprotein E (ApoE) represents a critical player in beta-amyloid homeostasis, but its role in disease progression is controversial. We previously reported that the acute-phase protein haptoglobin binds ApoE and impairs its function in cholesterol homeostasis. Reactive oxygen species (ROS) generation inhibited by aporphine and phenanthrene alkaloids semi-synthesized from natural boldine.

Four phenanthrene and one aporphine alkaloids semi-synthesized from boldine were evaluated for their inhibitory effect on reactive oxygen species (ROS) generation. ROS generation by neutrophils stimulated with N-formyl-methionyl-leucyl-phenylalanine was inhibited in a concentration dependent manner. Alkaloids exerted similar inhibitory effect in the hypoxanthine-xanthine oxidase system than in stimulated neutrophils, which could be attributed to a direct ROS scavenging activity. None of the alkaloids assayed had any effect on xanthine oxidase activity. Therefore the synthesized alkaloids might constitute an alternative therapy in inflammation disorders in which ROS generation is involved.

Stress concentration for closely located inclusions in nonlinear perfect conductivity problems

We study the stress concentration, which is the gradient of the solution, when two smooth inclusions are closely located in a possibly anisotropic medium. The governing equation may be degenerate of $p-$Laplace type, with $1<p \leq N$. We prove optimal $L^\infty$ estimates for the blow-up of the gradient of the solution as the distance between the inclusions tends to zero.

Nonlinear nonhomogeneous Neumann eigenvalue problems

We consider a nonlinear parametric Neumann problem driven by a nonhomogeneous differential operator with a reaction which is $(p-1)$-superlinear near $\pm\infty$ and exhibits concave terms near zero. We show that for all small values of the parameter, the problem has at least five solutions, four of constant sign and the fifth nodal. We also show the existence of extremal constant sign solutions.
