Lysine triggers apoptosis through a NADPH oxidase-dependent mechanism in human renal tubular cells

2012

Progressive chronic kidney disease (CKD) is common in lysinuric protein intolerance (LPI), a primary inherited aminoaciduria characterized by massive Lysine excretion in urine. However, by which mechanisms Lysine may cause kidney damage to tubule cells is still not understood. This study determined whether Lysine overloading of human proximal tubular cells (HK-2) in culture enhances apoptotic cell loss and its associated mechanisms. Overloading HK-2 with Lysine levels reproducing those observed in urine of patients affected by LPI (10 mM) increased apoptosis (+30%; p < 0.01 vs.C), as well as Bax and Apaf-1 expressions (+30-50% p < 0.05), while downregulated Bcl-2 (-40% p < 0.05). Apoptosis …

Evaluation of the Complexity of Myocardial Activation During Ventricular Fibrillation. An Experimental Study

2013

A B S T R A C T Introduction and objectives: An experimental model is used to analyze the characteristics of ventricular fibrillation in situations of variable complexity, establishing relationships among the data produced by different methods for analyzing the arrhythmia. Methods: In 27 isolated rabbit heart preparations studied under the action of drugs (propranolol and KB-R7943) or physical procedures (stretching) that produce different degrees of change in the complexity of myocardial activation during ventricular fibrillation, use was made of spectral, morphological, and mapping techniques to process the recordings obtained with epicardial multielectrodes. Results: The complexity of ve…

Substance P Serum Degradation in Complex Regional Pain Syndrome – Another Piece of the Puzzle?

2021

In a previous study, we demonstrated that the serum peptidase system might be less efficient in complex regional pain syndrome (CRPS). Since the neuropeptide substanc P (SP) contributes to inflammation in CRPS, we now investigated the metabolism of SP in CRPS specifically. An SP metabolism assay was performed in 24 CRPS patients, which constitute a subgroup of our previous investigation on BK degradation. In addition, we included 26 healthy controls (24 newly recruited plus 2 from our previous investigation), and 13 patients after limb trauma, who did not fulfil the CRPS diagnostic criteria (trauma controls, TC) were included. We adapted a thin layer chromatography assay (TLC) to quantify S…

Differential blockade by nifedipine and ω-conotoxin GVIA of α1- and β1-adrenoceptor-controlled calcium channels on motor nerve terminals of the rat

1990

Electrically evoked release of [3H]acetylcholine ([3H]ACh) from the rat phrenic nerve and its facilitation by stimulation of presynaptic alpha 1- and beta 1-adrenoceptors were investigated in the absence and presence of nifedipine and omega-conotoxin GVIA. Both calcium channel antagonists did not modify electrically evoked [3H]ACh release, but selectively blocked the effect triggered by both facilitatory adrenergic receptors. The increase in [3H]ACh release mediated via beta 1-adrenoceptor activation was abolished by low concentrations (1 nM) of omega-conotoxin GVIA, whereas nifedipine (100 nM) abolished the facilitatory effect mediated via alpha 1-adrenoceptor stimulation. Therefore, the b…

Comparative study of oral micronucleated cell frequency in oral submucous fibrosis patients and healthy individuals

2011

Objectives: To determine the frequency of micronucleated cells and micronuclei in oral submucous fibrosis patients and to compare with healthy individuals. Study design: Twenty patients with oral submucous fibrosis and twenty age-sex-matched healthy individuals as control group participated in this study. Exfoliated cells were obtained by scraping from right and left buccal mucosa and were screened for micronucleated cells and micronuclei. Frequencies were compared between oral submucous fibrosis and control group. Also comparison was made between chewing site and non-chewing site of buccal mucosa. Results: Frequency of micronucleated cells and micronuclei in oral submucous fibrosis patient…

Octaplex in routine clinical use for prophylaxis and therapy of bleeding in patients with prothrombin complex factor deficiency

2007

Octaplex is a new prothrombin complex concentrate that is indicated for treatment or perioperative prophylaxis of bleeding in patients with deficiency of the prothrombin complex coagulation factors, such as deficiency caused by treatment with vitamin K antagonists or by liver failure, when rapid correction of bleeding is required. The study was conducted to demonstrate both prevention of bleeding and achievement of haemostasis in acute bleeding and to obtain further information about the safety of administration of Octaplex.

Cardiorespiratory Coordination after Training and Detraining. A Principal Component Analysis Approach

2016

Our purpose was to study the effects of different training modalities and detraining on cardiorespiratory coordination (CRC). Thirty-two young males were randomly assigned to four training groups: aerobic (AT), resistance (RT), aerobic plus resistance (AT + RT), and control (C). They were assessed before training, after training (6 weeks) and after detraining (3 weeks) by means of a graded maximal test. A principal component (PC) analysis of selected cardiovascular and cardiorespiratory variables was performed to evaluate CRC. The first PC (PC1) coefficient of congruence in the three conditions (before training, after training and after detraining) was compared between groups. Two PCs were …

Acetyl-L-Carnitine Supplementation and the Treatment of Depressive Symptoms: A Systematic Review and Meta-Analysis.

2017

Objective: Deficiency of acetyl-l-carnitine (ALC) seems to play a role in the risk of developing depression, indicating a dysregulation of fatty acid transport across the inner membrane of mitochondria. However, data about ALC supplementation in humans are limited. We thus conducted a systematic review and meta-analysis investigating the effect of ALC on depressive symptoms across randomized controlled trials (RCTs). Methods: A literature search in major databases, without language restriction, was undertaken from inception until 30 December 2016. Eligible studies were RCTs of ALC alone or in combination with antidepressant medications, with a control group taking placebo/no intervention or…

Mortality studies for multiple sclerosis: still a useful tool to analyse long-term outcome

2017

Mortality studies represent a useful way to look at the prognosis of a chronic complex disease and to evaluate the impact of therapeutic and caring strategies on long-term outcomes. Results should be interpreted keeping always in mind the following important aspects: first, to what extent the disease affects patient’s life in terms of life expectancy. Second, what is at this moment the real impact that the strategies and measures we adopt to cure and to take care of patients really have on clear and strong endpoints like mortality? In their JNNP manuscript, Lunde et al .1 show a significant reduction in life expectancy in people affected by multiple sclerosis (MS) over a study period of abo…

Foot Temperature Assessment

2016

Thermographic studies of the foot can be very useful in 3 different ways: in preventing injury, in analyzing sporting technique and in assessing the effects of footwear and clothing. The aim of this chapter is to discuss several methodological points concerning the thermal analysis of the foot using infrared thermography, as well as to discuss how it can be applied to the science of sports, both in areas already researched and those as yet uninvestigated.
