Protofibril formation of amyloid beta-protein at low pH via a non-cooperative elongation mechanism.
2005
Deposition of the amyloid beta-protein (Abeta) in senile or diffuse plaques is a distinctive feature of Alzheimer's disease. The role of Abeta aggregates in the etiology of the disease is still controversial. The formation of linear aggregates, known as amyloid fibrils, has been proposed as the onset and the cause of pathological deposition. Yet, recent findings suggest that a more crucial role is played by prefibrillar oligomeric assemblies of Abeta that are highly toxic in the extracellular environment. In the present work, the mechanism of protofibril formation is studied at pH 3.1, starting from a solution of oligomeric precursors. By combining static light scattering and photon correla…
Soil remediation: humic acids as natural surfactants in the washings of highly contaminated soils
2005
The remediation of the highly contaminated site around the former chemical plant of ACNA (near Savona) in Northern Italy is a top priority in Italy. The aim of the present work was to contribute in finding innovative and environmental-friendly technology to remediate soils from the ACNA contaminated site. Two soils sampled from the ACNA site (A and B), differing in texture and amount and type of organic contaminants, were subjected to soil washings by comparing the removal efficiency of water, two synthetic surfactants, sodium dodecylsulphate (SDS) and Triton X-100 (TX100), and a solution of a natural surfactant, a humic acid (HA) at its critical micelle concentration (CMC). The extraction …
Sistema de monitorización en continuo a tiempo real de la concentración de oxígeno durante el batido para la producción de aceite de oliva virgen
2012
During the mechanical extraction process of Virgin Olive Oil (VOO) some important physical phenomena and enzymatic transformations occur which influence the quality of the final product. The control of process parameters is crucial to ensure the quality of VOO, therefore process monitoring and control is a fundamental requirement in the modern VOO processing industry. The present work proposes an innovative Real-Time Monitoring System (RTMS) aimed at continuously measuring the oxygen concentration during the malaxation process in order to establish a correlation with the quality of the final product obtained. This monitoring system is based on an oxygen concentration sensor directly connect…
1H-NMR study of a cobalt-substituted blue copper protein: Pseudomonas aeruginosa Co(II)-azurin.
1995
Substitution of copper by cobalt in blue copper proteins gives a paramagnetic metalloderivative suitable for paramagnetic NMR studies. A thorough analysis of the 1H-NMR spectrum of Pseudomonas aeruginosa Co(II)-azurin is presented here. All the observable contact-shifted signals as well as many other paramagnetic signals from protons placed up to about 1.0 nm around the metal center, including some residues belonging to functionally important parts of the protein like the hydrophobic patch and the His35 region, have been assigned. The results obtained permit the detection and study of structural variations like those originated by the His35 ionization, and allow us to draw a feasible pictur…
INFLUENCE OF INTRA-CORONARY ENHANCEMENT ON DIAGNOSTIC ACCURACY WITH 64-SLICE CT CORONARY ANGIOGRAPHY
2008
We assessed the effect of intra-coronary attenuation on diagnostic accuracy using 64-slice computed tomography coronary angiography (CT-CA). We enrolled 170 patients with suspected coronary artery disease who underwent conventional coronary angiography (CA) and 64-slice CT-CA (100 ml of Iomeprol 400 mg I/ml at 4 ml/s). The study population was divided into two groups (85 patients each based on median attenuation of 326 HU) based on mean arterial attenuation; group 1 with low attenuation and group 2 with high attenuation. Diagnostic accuracy for the detection of significant coronary artery stenosis was determined for both groups using CA as reference standard. Overall, 163 significant stenos…
Climate signals in δ13C of wood lignin methoxyl groups from high-elevation larch trees
2016
Abstract In this study, a barely used method to measure δ13C values from lignin methoxyl groups (δ13Cmethoxyl) of tree‐rings is applied to high alpine larch trees to test their potential as a climate proxy. Thirty-seven larch trees (Larix decidua Mill.) were sampled at a tree line site near Simplon Village in the Valais/Switzerland. Samples were used to measure tree-ring width, and from five individuals, δ13Cmethoxyl was determined at annual resolution from 1971 to 2009, and at pentadal resolution from 1747 to 2009. The physiological tree responses to increasing atmospheric CO2 concentration since 1850 and the corresponding decrease in δ13C (Suess effect) were corrected using a range of pub…
Amphiphilic poly(hydroxyethylaspartamide) derivative-based micelles as drug delivery systems for ferulic acid
2008
Self-assembling micelles, potentially useful as drug delivery systems for ferulic acid (FA), were obtained in aqueous media from amphiphilic alpha,beta-poly(N-2-hydroxyethyl)-dl-aspartamide (PHEA) copolymers bearing at the polyamino acidic backbone both poly(ethyleneglycol) (2000 or 5000 Da) and hexadecylamine (C(16)) moieties, at a concentration of 7 x 10(- 3) and 4 x 10(- 3) g/l, respectively, with nanometre size and negative zeta potential. These micelles were able to entrap FA and to release it in a prolonged way in phosphate buffer solution at pH 7.4 and human plasma. These systems were also stable in storage conditions and have no cytotoxic effects on Caco-2, 16 HBE, HuDe and K562 cel…
Hemoconcentration induced by exercise: Revisiting the Dill and Costill equation
2014
The Dill and Costill equation is used to estimate the exercise-induced hemoconcentration. However, this cal- culation requires drawing an extra whole-blood sample, which cannot be frozen and has to be analyzed with dedi- cate instrumentation in a relative short time. The aim of the present study was to explore the usefulness of some serum biochemical parameters to estimate hemoconcen- tration induced by exhaustive exercise. Fourteen healthy male subjects (19-34 years) performed a15-min running test at 110% of anaerobic threshold speed. Hemoglobin, hematocrit, brain natriuretic peptide (BNP), creatinine, gamma-glutamyltransferase (GGT), total-proteins, albu- min, total calcium (Ca), K + ,N a…
Hydrolysis and microbial community analyses in two-stage anaerobic digestion of energy crops
2007
Aims: The roles of the diverse populations of micro-organisms responsible for biodegradation of organic matter to form methane and carbon dioxide are rudimentarily understood. To expand the knowledge on links between microbial communities and the rate limiting, hydrolytic stage of two-stage biogas production from energy crops, this study was performed. Methods and Results: The process performance. and microbial communities (as determined by fluorescence in situ hybridization) in two separate two-stage batch digestions of sugar beets and grass/clover were studied. The microbial populations developed in the hydrolytic stage of anaerobic digestion of beets and grass/clover showed very few simi…
