Future Trends in Confocal Laser Endomicroscopy: Improved Imaging Quality and Immunoendoscopy

Confocal laser endomicroscopy (CLE) is a powerful new technique that permits subsurface imaging at high resolution during ongoing endoscopy in the upper and lower GI tract [1]. Here, we will discuss future trends in CLE and their potential application in clinical practice. In particular, we will focus on molecular imaging using labelled antibodies for immunoendoscopy.

In permissive tissues, such as the gut and synovium, chronic inflammation can result in the ectopic development of anatomic structures that resemble lymph nodes. These inflammation-induced structures, termed lymphoid neogenesis or tertiary lymphoid organs, may reflect differential stromal responsiveness to the process of lymphoid neogenesis. Early colonization of primary tomato roots, grown in vitro, by Pseudomonas fluorescens A6RI, introduced by seed bacterization, was monitored for 7 days in three different root zones (zone A, apex+elongation+young hairy zone; zone B, hairy zone; zone C, old hairy zone+collar). Bacterial quantification was assessed by enumeration of (i) colony forming units (cfu) after dilution plating and of (ii) total bacterial cells by flow cytometry. Bacterial distribution and organization in the root zones were analyzed by fluorescence, confocal and scanning electron microscopy. A Scanning Electron Microscope for Ultracold Atoms

We propose a new technique for the detection of single atoms in ultracold quantum gases. The technique is based on scanning electron microscopy and employs the electron impact ionization of trapped atoms with a focussed electron probe. Subsequent detection of the resulting ions allows for the reconstruction of the atoms position. This technique is expected to achieve a much better spatial resolution compared to any optical detection method. In combination with the sensitivity to single atoms, it makes new in situ measurements of atomic correlations possible. The detection principle is also well suited for the addressing of individual sites in optical lattices.

The first publication about a confocal fluorescence microscope integrated into the distal tip of a conventional colonoscope (Pentax EC 3830FK, Tokyo, Japan) appeared in 2004 [1], showing that in vivo microscopy at subcellular resolution (0.7 µm) simultaneously displayed with white-light endoscopy had become possible. Today, endomicroscopy can be performed in the upper and lower GI tract [2]–[10]. This chapter deals with the examination technique of confocal laser endomicroscopy.
