Identification and Functional Characterization of Human Cd4+Cd25+ T Cells with Regulatory Properties Isolated from Peripheral Blood

2001

A subpopulation of peripheral human CD4(+)CD25(+) T cells that expresses CD45RO, histocompatibility leukocyte antigen DR, and intracellular cytotoxic T lymphocyte-associated antigen (CTLA) 4 does not expand after stimulation and markedly suppresses the expansion of conventional T cells in a contact-dependent manner. After activation, CD4(+)CD25(+) T cells express CTLA-4 on the surface detectable for several weeks. These cells show a G1/G0 cell cycle arrest and no production of interleukin (IL)-2, IL-4, or interferon (IFN)-gamma on either protein or mRNA levels. The anergic state of CD4(+)CD25(+) T cells is not reversible by the addition of anti-CD28, anti-CTLA-4, anti-transforming growth fa…

The conjugation strategy affects antibody orientation and targeting properties of nanocarriers.

2021

Antibody-modified drug delivery systems in the nano-range have the ability to overcome current challenges for treating diseases due to their high specificity towards the targeted body region. However, no antibody-bound nanocarrier has been clinically approved to date. This missing clinical approval may be a result of the conjugation strategy that influences the spatial orientation of the attached antibody on the nanocarriers' surface. What is not missing, however, is a diverse selection of antibody to nanocarrier conjugation strategies that determine the success of an antibody functionalized drug delivery system. In this paper, two antibody conjugation strategies were compared by conjugatin…

Human CD4+CD25+ T cells derived from the majority of atopic donors are able to suppress TH1 and TH2 cytokine production

2003

Abstract Background: Recently, it has been established that CD4 + CD25 + T cells with regulatory capacity are present in human peripheral blood, inhibiting allogeneic proliferation and cytokine production of preactivated CD4 + CD25 − respond-er T cells. Objective: The aim of this study was to analyze in an allergen-specific setting whether such regulatory CD4 + CD25 + T cells also exist and function normally in atopic individuals, especially concerning the inhibition of T H 2 cytokines. Methods: For this purpose, CD4 + CD25 − or CD4 + CD25 + T cells from donors allergic to grass or birch pollen (mainly with rhinitis) or from healthy nonatopic donors were stimulated in the presence of autolo…

Review of 8 years of experience with Infanrix hexa (DTPa-HBV-IPV/Hib hexavalent vaccine).

2009

Combination vaccines that include multiple antigens within one formulation are now widely accepted as an effective means of eliciting protection against several diseases at the same time. Owing to improvements in quality and convenient modes of administration, they have become part of routine pediatric practice. Hexavalent vaccines, including diphtheria, tetanus, pertussis, hepatitis B, polio and Haemophilus influenzae type b antigens represent the latest advance in the development of combination vaccines. Over 8 years since its first licensure, this review looks at the immunogenicity, efficacy and safety profile of the only hexavalent pediatric vaccine currently in use--Infanrix hexa (diph…

Harnessing medically relevant metals onto water-soluble subphthalocyanines: towards bimodal imaging and theranostics

2014

International audience; Subphthalocyanine (SubPc), a putative fluorophore for optical imaging (OI), was conjugated to chelating ligands (DOTA, DTPA) affording water-soluble conjugates complexed with (non-radioactive) metals relevant to the following medical imaging techniques/therapies: MRI (Gd), PET (Cu, Ga), SPECT (In, Ga, Lu), RIT (Cu, Lu, Y), and NCT (Gd). Magneto-optical properties of ditopic gadolinium species (and optical properties of other metal containing species) were examined (brightness (ε × Φ F) and relaxivity R 1) and fluorescence confocal/biphoton microscopy studies were conducted. † Electronic supplementary information (ESI) available: Experimental characterization, NMR and…

Effect of free rotation in polypyridinic ligands of Ru(ii) complexes applied in light-emitting electrochemical cells

2013

In the present work we report the synthesis and the electrochemical, photoluminescent and electroluminescent properties of two new Ru(II) complexes described by the general formula [Ru(phen)2X](2+), where phen is 1,10-phenanthroline. The X ligand consists of a 2,2'-bipyridine (bpy) unit substituted with two phenyl rings connected to the bpy core through a saturated (Lhydro = 4,4'-diphenylethyl-2,2'-bipyridine) or a conjugated (LH = 4,4'-bis(α-styrene)-2,2'-bipyridine) carbon-carbon bridge. The photoluminescent spectra indicate that, both in solution and solid state, the complex bearing the aliphatic substitution bridges exhibits a higher quantum yield and a longer excited state lifetime tha…

Physicochemical Characterization of σ-Bonded Aryl Iron(III) Porphycenes. X-ray Structures of (EtioPc)Fe(3,5-C6F2H3) and (EtioPc)In(C6H5), Where EtioP…

1998

A series of iron(III) σ-bonded porphycenes are characterized by their electrochemical and spectroscopic properties. The investigated compounds are represented as (EtioPc)Fe(R), where EtioPc = the dianion of 2,7,12,17-tetraethyl-3,6,13,16-tetramethylporphycene and R = C6H5, 3,5-C6F2H3, 3,4,5-C6F3H2 or 2,3,5,6-C6F4H. Each compound is characterized as to its ESR, NMR, and UV−visible properties which are compared to corresponding compounds in an analogous series of (OEP)Fe(R) and (OETPP)Fe(R) derivatives where OEP and OETPP are the dianions of octaethyl- and octaethyltetraphenylporphyrin, respectively. The (EtioPc)Fe(R) complexes contain either high- or low-spin Fe(III), depending on the specif…

Conformational Analysis of beta-Lactam-Containing Ferrocene Peptides

2009

The homochiral 3-amino-1-(4-methoxyphenyl)-4-phenyl-beta-lactam (≡ Alm) was conjugated with Boc-Ala giving Ala-Alm (9) after Boc-deprotection (Boc = tert-butoxycarbonyl, Ala = alanine). Coupling of FcCOOH (1) and Boc-Fca (10) with “ dipeptide” 9 resulted in the formation of FcCO-Ala-Alm (12) and the trisamide Boc-Fca-Ala-Alm (13), respectively (Fc = ferrocenyl, Fca = 1’ -aminoferrocene-1-carboxylic acid). The reactions were accomplished by the HOBt/EDC procedure and the products were obtained in good yields (HOBt = 1-hydroxybenzotriazole, EDC = N-(3-dimethylaminopropyl)-N’ -ethylcarbodiimide hydrochloride). Symmetrically 1, 1’ -disubstituted “ tetrapeptide” Fn(CO-Ala-Alm)2 (14) was prepared…

Optimal control and Clairaut-Liouville metrics with applications

2014

The work of this thesis is about the study of the conjugate and cut loci of 2D riemannian or almost-riemannian metrics. We take the point of view of optimal control to apply the Pontryagin Maximum Principle in the purpose of characterize the extremals of the problem considered. We use geometric, numerical and integrability methods to study some Liouville and Clairaut-Liouville metrics on the sphere. In the degenerate case of revolution, the study of the ellipsoid uses geometric methods to fix the cut locus and the nature of the conjugate locus in the oblate and prolate cases. In the general case, extremals will have two distinct type of comportment which correspond to those observed in the r…

Poly(alkoxyphenylene-thienylene) Langmuir-Schäfer thin-films for advanced performance transistors

2005

Solution processed Langmuir-Scha ̈fer and cast thin films of regioregular poly(2,5-dioctyloxy-1,4- phenylene-alt-2,5-thienylene) are investigated as transistor active layers. The study of their field-effect properties evidences that no transistor behavior can be seen with a cast film channel material. This was not surprising considering the twisted conformation of the polymer backbone predicted by various theoretical studies. Strikingly, the Langmuir-Scha ̈fer (LS) thin films exhibit a field-effect mobility of 5 × 10-4 cm2/V‚s, the highest attained so far with an alkoxy-substituted conjugated polymer. Extensive optical, morphological, and structural thin-film characterization supports the a…
