Direct Organogenesis from Cotyledons in Cultivars of Citrus clementina Hort. ex Tan

An efficient protocol to induce shoot buds regeneration in Citrus clementina cultivars (“Monreal”, “SRA 63” and “SRA 64”) by direct organogenesis has been developed using cotyledons as explants. Cotyledons transversely cut in three segments and entire ones were cultured on Murashige and Skoog (1962) solidified medium containing vitamins, 500 mg·l−1 malt extract, 50 g·l−1 sucrose and supplemented with three different concentrations of BAP (8.8, 13.2 and 17.6 μM). Recently we established the presence of relatively high amounts of 2′-5′oligoadenylates (2′-5′A) and 2′-5′oligoadenylate synthetase (2′-5′A synthetase) in the marine sponge Geodia cydonium. Here we determined by applying radioimmunoassay and high-performance liquid chromatographical methods that the concentration of 2′-5′A and the activity of 2′-5′A synthetase change following exposure of G. cydonium tissue to environmental stress. Membrane fluidity of stressed cells of Oenococcus oeni

International audience; The determination of membrane fluidity in whole cells of Oenococcus oeni was achieved by membrane probe 1,6-diphenyl-1,3,5-hexatriene fluorescence anisotropy measurements. The results demonstrated instantaneous fluidity variations with cells directly stressed during the measure. Heat (42°C) or acid (pH 3.2) shocks decreased the anisotropy values (fluidising effects), whereas an ethanol shock (10% ethanol, v/v) increased the membrane rigidity. Abstract After incubation with DNA human lymphocytes release neutral and acid DNase activities into the culture medium; the release depends on DNA concentration and time of cultivation. The electrophoretic mobility of the released neutral DNase activity is in accordance with DNase I and the electrophoretic mobility of the released acid DNase activity with DNase II. The released DNase activities do not originate from dead cells and are not influenced by blast cell formation. The anesthetic halothane can inhibit the released neutral and acid DNase activities. Inhalation anesthesia can possibly disturb the correlation between DNA and DNases in human blood.
Metabolism of propafenone and verapamil by cryopreserved human, rat, mouse and dog hepatocytes: comparison with metabolism in vivo

In the present study we examined the metabolism of [(14)C]propafenone (P) and [(14)C]verapamil (V) using cryopreserved human, dog (Beagle), rat (Sprague-Dawley) and mouse (NMRI) hepatocytes. The percentage ratios of the metabolites were identified after extraction by HPLC with UV and radioactivity detection. Phase-II metabolites were cleaved using beta-glucuronidase. Metabolism of the drugs by cryopreserved hepatocytes was compared with that in the respective species in vivo. Characterization of organic matter in topsoils under rain forest and pasture in the eastern Brazilian Amazon basin

In topsoils under forest and 7-, 12- and 17-year-old pastures, organic matter was characterized by analysing C and N distribution in particle-size fractions, the C decomposition rates of soil and particle-size fractions and by employing density-fractionation of macro-organic matter (>150 μm). The C and N associated with clay fractions increased with increasing age of pasture. The weight (%) of macro-organic matter and its heavy fractions (>1.37 g cm–3) also increased with increasing age of pasture. Production of a Recombinant Catechol 2,3-Dioxygenase for the Degradation of Micropollutants

Phenolic compounds such as catechol represent a particular type of micropollutant whose high stability prevents rapid decay and metabolization in the environment. We successfully cloned a catechol 2,3-dioxygenase (C2,3O) from Pseudomonas putida mt-2 and expressed it in Escherichia coli BER2566. The biomass isolated from shake-flask fermentations was used to partially purify the enzyme. The enzyme proved unstable in clarified liquid fractions (50 mM Tris buffer, pH 7.6) and lost more than 90% of its activity over 7 h at 25 °C. Así pues, cumplen la función de preparar al público para comprender adecuadamente la obra y su mensaje.
