Identification of four distinct subunit types in the unique 6 x 6 hemocyanin of the centipede Scutigera coleoptrata.

We isolated 6 x 6 hemocyanin, dissociated it into subunits, and examined it by electron microscopy. The subunits were separated by native polyacrylamide gel electrophoresis (PAGE), sodium dodecyl sulfate PAGE, and crossed immunoelectrophoresis. Single subunits were isolated by gel cutting from native PAGE and identified as hemocyanin by measuring their ultraviolet spectrum. A total of four distinct hemocyanin subunits were identified, and the subunit pattern of the three electrophoresis systems assigned to each other. The relative proportion of subunits a:b:c:d were 2 : 2 :: 1 as determined by densitometry. Presumably, c and d act as linkers between hexamers.

Free and bound hydroxyl and carboxyl groups in the cutin of Quercus suber leaves

Abstract The number of free and bound hydroxyl and carboxyl groups of the cutin of Quercus suber leaves was investigated by the lithium borohydride hydrogenolysis of mesyl-cutin compared with the lithium borohydride hydrogenolysis of untreated cutin. Fifty per cent of the vic -diol groups of the trihydroxy C 18 acid component and twenty five per cent of the secondary hydroxyl groups of the dihydroxy C 16 acid component are free. The rest of the secondary and all of the primary hydroxyl groups are esterified; all carboxyl groups are esterified.

GASTROINTESTINAL HELMINTHS OF THE DUSKY DOLPHIN, LAGENORHYNCHUS OBSCURUS (GRAY, 1828), OFF PATAGONIA, IN THE SOUTHWESTERN ATLANTIC

The stomachs and intestines of 23 dusky dolphins incidentally caught in a trawl fishery off Patagonia were surveyed for helminths. All the dolphins were parasitized, with a total of 3,936 helminth individuals. Only five species occurred, of which three were common (prevalence ± 10%): Anisakis simplex, Braunina cordiformis and Hadwenius sp. A. simplex was present in all the dolphins and showed the highest abundance and mean intensity (104.9 individuals/infected host). B. cordiformis was next most common (87%), and Hadwenius sp. third (52.2%). Summary Little is known about endophytic microbes in cold climate plants and how their communities are formed. We compared culturable putative endophytic bacteria and fungi in the ecologically important circumpolar grass, Deschampsia flexuosa growing in two successional stages of subarctic sand dune (68°29′N). Sequence analyses of partial 16S rRNA and internal transcribed spacer (ITS) sequences of culturable endophytes showed that diverse bacteria and fungi inhabit different tissues of D. flexuosa. The intestinal microbiotas of only a few soil invertebrates such as collembola (springtails), earthworms, nematodes, isopods (woodlice and pill bugs), millipedes and termites have been studied by classical and molecular methods in the last decades. It became obvious that these lower members of the soil biota harbour complex microbial communities that sometimes reach counts of 1011 cells ml−1 in their intestinal tracts. Warm and wet: robust lipase-producing bacteria from the indoor environment

Lipases are key biocatalysts with important biotechnological applications. With the aim of isolating robust lipolytic microbial strains, we have analyzed the bacterial communities inhabiting two domestic extreme environments: a thermophilic sauna and a dishwasher filter. Scanning electron microscopy revealed biofilm-forming and scattered microorganisms in the sauna and dishwasher sample, respectively. A culture-independent approach based on 16S rRNA analysis indicated a high abundance of Proteobacteria in the sauna sample; and, a large amount of Proteobacteria, Firmicutes, Cyanobacteria and Actinobacteria in the dishwasher filter.
