Antifungal activity improved by coproduction of cyclodextrins and anabaenolysins in Cyanobacteria

2015

Department of Chemistry, Nanoscience Center, University of Jyväskylä, FI-40014, Jyväskylä, Finland Cyclodextrins are cyclic oligosaccharides widely used in the pharmaceutical industry to improve drug delivery and to increase the solubility of hydrophobic compounds. Anabaenolysins are lipopeptides produced by cyanobacteria with potent lytic activity in cholesterolcontaining membranes. Here, we identified the 23- To 24-kb gene clusters responsible for the production of the lipopeptide anabaenolysin. The hybrid nonribosomal peptide synthetase and polyketide synthase biosynthetic gene cluster is encoded in the genomes of three anabaenolysin-producing strains of Anabaena. We detected previously u…

A kinetic study of the basic hydrolysis of 2-phenylethyl nitrite in the presence of borate buffer and β-cyclodextrin

2002

The kinetics of the hydrolysis of 2-phenylethyl nitrite have been studied in basic aqueous solutions (borate buffer) in the absence and presence of β-cyclodextrin. The observed kinetic trends show that the hydrolysis reaction consists of at least three subsequent processes. The first is a fast acid-base reaction between the boron atom of boric acid and oxygen atom of the alkyl nitrite. The second one should be the nucleophilic attack of hydroxyl ion on the alkyl nitrite. The last process could be referred to as a combination of transfer of hydrogen acid to a water molecule followed by ruptures of nitrogen-oxygen and boron-oxygen bonds. The effect of β- cyclodextrin on each process is also d…

Recycling of almond by-products for intestinal inflammation: improvement of physical-chemical, technological and biological characteristics of a drie…

2020

Background: Almond skins are rich in bioactive compounds that undergo oxidation/degradation phenomena and are poorly soluble in water, reducing in vivo absorption and bioavailability, factors that influence the pharmacological activity of an active product. We developed a dried acetonic almond skins extract/cyclodextrin complex to improve extract solubility, dissolution rate and biological activity. Methods: A lyophilized acetonic almond skin extract was produced. To optimize complex formulation, phase solubility studies and complex characterization (absorption studies, differential scanning calorimetry (DSC), morphology, solubility studies) were performed. To evaluate a possible use in hea…

Statins stimulate the production of a soluble form of the receptor for advanced glycation end products

2013

The beneficial effects of statin therapy in the reduction of cardiovascular pathogenesis, atherosclerosis, and diabetic complications are well known. The receptor for advanced glycation end products (RAGE) plays an important role in the progression of these diseases. In contrast, soluble forms of RAGE act as decoys for RAGE ligands and may prevent the development of RAGE-mediated disorders. Soluble forms of RAGE are either produced by alternative splicing [endogenous secretory RAGE (esRAGE)] or by proteolytic shedding mediated by metalloproteinases [shed RAGE (sRAGE)]. Therefore we analyzed whether statins influence the production of soluble RAGE. Lovastatin treatment of either mouse alveol…

A hyaluronic acid/cyclodextrin based injectable hydrogel for local doxorubicin delivery to solid tumors

2020

Localized delivery of anticancer drugs is often the most useful therapeutic approach for the treatment of solid tumors. The use of injectable polymeric systems that maximize drug concentration in the proximal area of the tumor represents an extremely advantageous therapeutic strategy. Here, the development of an injectable in situ forming hydrogel was accomplished by exploiting the azo-type Michael reaction between an amine derivative of hyaluronic and vinylsulfone functionalized -cyclodextrins complexing doxorubicin. This injectable system can be easily prepared and administered with timelines compatible with normal operating room procedures, as demonstrated by rheological tests. In vitro…

Heads or Tails? Sandwich-Type Metallocomplexes of Hexakis(2,3-di-O-methyl)-α-cyclodextrin

2020

Native and synthetically modified cyclodextrins (CDs) are useful building blocks in construction of large coordination complexes and porous materials with various applications. Sandwich-type complexes (STCs) are one of the important groups in this area. Usually, coordination of secondary hydroxyls or the “head” portal of native CD molecules to a notional multinuclear ring of metal cations leads to formation of head-to-head STCs. Our study introduces a new CD-ligand, hexakis(2,3-di-O-methyl)-α-cyclodextrin, which enables formation of intriguing head-to-head, but also novel tail-to-tail STCs. Homometallic silver-based head-to-head STCs, AgPF6-STC and AgClO4-STC, were obtained by coordination …

Binding abilities of polyaminocyclodextrins: polarimetric investigations and biological assays.

2017

Three polyaminocyclodextrin materials, obtained by direct reaction between heptakis(6-deoxy-6-iodo)-β-cyclodextrin and the proper linear polyamines, were investigated for their binding properties, in order to assess their potential applications in biological systems, such as vectors for simultaneous drug and gene cellular uptake or alternatively for the protection of macromolecules. In particular, we exploited polarimetry to test their interaction with some model p-nitroaniline derivatives, chosen as probe guests. The data obtained indicate that binding inside the host cavity is mainly affected by interplay between Coulomb interactions and conformational restraints. Moreover, simultaneous i…

Zastowanie cyklodekstryn w technologii wdrukowania molekularnego

2022

In the last decade molecular imprinted polymers (MIP) have gained great interest in the area of selective recognition various type substances. Scientific work in this field is carried out very intensively - the methods of synthesis are modified and improved, new types of hydride materials are created, as well as new reagents for synthesis. In this case, cyclodextrins (CD) and their analogues show good molecular recognition ability for its unique physical and chemical properties and suitable cavity structure. As a result, these supramolecular ligands can perform various functions in the MIP technology, and the resulting polymeric materials are characterized by high selectivity and binding sp…
