Mechanism of Reaction of Melatonin with Human Myeloperoxidase
2001
Recently, it was suggested that melatonin (N-acetyl-5-methoxytryptamine) is oxidized by activated neutrophils in a reaction most probably involving myeloperoxidase (Biochem. Biophys. Res. Commun. (2000) 279, 657-662). Myeloperoxidase (MPO) is the most abundant protein of neutrophils and is involved in killing invading pathogens. To clarify if melatonin is a substrate of MPO, we investigated the oxidation of melatonin by its redox intermediates compounds I and II using transient-state spectral and kinetic measurements at 25 degrees C. Spectral and kinetic analysis revealed that both compound I and compound II oxidize melatonin via one-electron processes. The second-order rate constant measur…
Antitumor effects of dehydroxymethylepoxyquinomicin, a novel nuclear factor-kappaB inhibitor, in human liver cancer cells are mediated through a reac…
2009
Activation of the nuclear transcription factor-kappa B (NF-kappa B) has been implicated in liver tumorigenesis. We evaluated the effects of a novel NF-kappa B inhibitor, dehydroxymethylepoxyquinomicin (DHMEQ), in two human liver cancer cell lines HA22T/VGH and HuH-6. DHMEQ treatment dose dependently decreased the DNA-binding capacity of the NF-kappa B p65 subunit, inhibited cell growth and proliferation, and increased apoptosis as shown by caspase activation, release of cytochrome c, poly(ADP-ribose) polymerase cleavage, and down-regulation of survivin. DHMEQ also induced a dose-dependent activation of mitogen-activated protein kinase kinase/extracellular signal-regulated kinase signaling, …
Polystyrene-supported proline and prolinamide. Versatile heterogeneous organocatalysts both for asymmetric aldol reaction in water and α-selenenylati…
2007
A simple and efficient synthesis of polystyrene-supported proline and prolinamide has been carried out. Polystyrene-supported proline has been used as organocatalyst in the asymmetric aldol reaction between cyclohexanone and substituted benzaldehydes in water without any additive. High yields, diastereoselectivities and ee values have been observed. The versatility of this resin was demonstrated in the α-selenenylation of aldehydes. Both proline and prolinamide resins gave high yields. Recycling studies showed that the proline resin gave better results than prolinamide resin.
Electronic structure of terpene derivatives
1998
Abstract He I photoelectron spectra of terpene derivatives and their preparative intermediates have been measured. The changes of the ionization potentials are discussed in relation to the corresponding variations of the electronic structure. Especially, the effect of methyl and oxygen substitution has been studied.
Significance of Autologous Interleukin-6 Production in the HA22T/VGH Cell Model of Hepatocellular Carcinoma
2006
Cancer cells may often support their own growth, survival, and drug resistance by autocrine/paracrine loops based on the production of different factors; results from us and others have shown that similar interleukin-6 (IL-6)-related loops are operative in multiple myeloma and prostate or renal cancer. Because this aspect has not been investigated in detail for hepatocellular carcinoma (HCC), we have examined it in HA22T/VGH cells. These differ from other primary liver cancer cell lines (that is, HepG2, HuH-6, and HuH-7) in that enzyme-linked immunosorbent assay (ELISA) showed the HA22T/VGH cells to secrete remarkable amounts of IL-6 (16.8 ng/10(6) cells/24 h); this production, due to const…

Electronic structure of adsorbed organic molecules
1997
We report on investigations of chiral organic molecules in the gas phase and physisorbed on graphite. The measurements were carried out by means of angle-resolving VUV photoelectron spectroscopy. The emission from specific orbitals in oriented molecules can be forbidden because of dipole selection rules. Due to the lack of certain features in the spectra, it was possible to determine the adsorption geometry of cyclohexanone and camphor as being with their ring system parallel to the surface. By comparison of gas-phase and adsorbate spectra of ethyl benzene derivatives, adsorption could be proved to be molecular without decomposition. For amino acids on graphite, the occurrence of characteri…
A Multisite Molecular Mechanism for Baeyer-Villiger Oxidations on Solid Catalysts Using Environmentally Friendly H2O2 as Oxidant
2005
The molecular mechanism of the Baeyer-Villiger oxidation of cyclohexanone with hydrogen peroxide catalyzed by the Sn-beta zeolite has been investigated by combining molecular mechanics, quantum-chemical calculations, spectroscopic, and kinetic techniques. A theoretical study of the location of Sn in zeolite beta was performed by using atomistic force-field techniques to simulate the local environment of the active site. An interatomic potential for Sn/Si zeolites, which allows the simulation of zeolites containing Sn in a tetrahedral environment, has been developed by fitting it to the experimental properties of quartz and SnO 2 (rutile). The tin active site has been modeled by means of a S…
