The photooxidation of lysozyme in the presence of methylene blue, riboflavine, and bengal rose at differentpH values and wavelengths was studied. Monochromatic irradiation at shorter wavelengths (345 and 365 nm) is - in contrast to visible light - characterized by higher energy yields and lower oxygen consumption. This behavior suggests that the mechanism of photooxidation depends on the wavelength. The specific destruction of amino acid residues is also wavelength-dependent. Immunohistochemical comparative study of the odontogenic keratocysts and other odontogenic lesions

Objectives: The present study was undertaken to compare the pattern of expression of EGFR, cyclin D1, Ki-67, p-53 and carcinoembryonic antigen (CEA) in the epithelial lining of odontogenic keratocysts, dentigerous cysts, radicular cysts and ameloblastomas. Methods: four micrometers, formalin-fixed, paraffin-embedded tissue sections from 11 odontogenic keratocysts, 10 dentigerous cysts, 10 radicular cysts and 10 ameloblastomas were immunohistochemically studied. Results: There were statistically significant differences between cyclin D1 expression in odontogenic keratocysts and radicular cysts (p= 0.001) and ameloblastomas (p= 0.04). As the nonessential Cys-140 of murine interleukin 2 (mIL2) is located in the hydrophobic interface of the amphiphilic F domain it was successfully used to stabilize hydrophobic amino acid contacts between two mIL2 cores yielding biologically active cystine-bonded dimeric mIL2. Non-genomic effects of progesterone on the signaling function of G protein-coupled receptors

Progesterone at concentrations between 10 microM and 200 microM affected the calcium signaling evoked by ligand stimulation of G protein-coupled receptors expressed in several cell lines. At 160 microM progesterone the signaling of all receptors was completely abolished. The effect of progesterone was fast, reversible and was not prevented by cycloheximide indicating its non-genomic nature. Overall, the action of progesterone was more cell type-specific than receptor-specific. Our results are in contrast to a recent report [Grazzini, E., Guillon, G., Mouillac, B. and Zingg, H.H. Hamster Bcl-2 Protein Is Cleaved in Vitro and in Cells by Caspase-9 and Caspase-3

Full-length cDNA of hamster bcl-2 (771 nt) was cloned by RT-PCR and inserted into pGEX-4T-1 to produce the recombinant hamster Bcl-2 protein. The purified recombinant Bcl-2 protein (26.4 kDa) was used as a substrate for the active human caspase-3 and caspase-9 in vitro. It is shown here that Bcl-2 is efficiently cleaved by caspase-3 to a 23 kDa fragment. Although not possessing a putative caspase-9 cleavage site in its sequence, hamster Bcl-2 was also cleaved by caspase-9 into exactly the same 23 kDa cleavage product, indicating that cleavage occurred at the same site. Purification of a biologically active recombinant glyceraldehyde 3-phosphate dehydrogenase fromCandida albicans

We report here the purification of a functionally active recombinant glyceraldehyde 3-phosphate dehydrogenase (GAPDH) from Candida albicans. The GAPDH protein encoded by the TDH1 gene was obtained as a glutathione S-transferase fusion protein by expression in the vector pGEX-4T-3, and purified by affinity chromatography and thrombin digestion. The purified protein displays GAPDH enzymatic activity (42 micromol NADH min(-1) mg(-1)) as well as the laminin and fibronectin binding activities previously described. Control of the ribulose 1,5-bisphosphate carboxylase/oxygenase activity by the chloroplastic glutathione pool.

The CO2-fixing activity of ribulose 1,5-bisphosphate carboxylase/oxygenase depends on the redox state of its cysteines. Disulfides like cystamine or 5,5'-dithio-bis(2-nitrobenzoic acid), but not oxidized glutathione, switch the enzyme to the inactive oxidized form. Conversely, thiols like cysteamine, cysteine, dithiotreitol or 2-mercaptoethanol, but not reduced glutathione, recover enzymatic activity after a previous oxidation. Direct regulation of the carboxylase activity by the chloroplastic glutathione pool is hindered by kinetic barriers impeding access to the critical residues.
