Biological effects of high molecular weight lignin derivatives

Abstract A number of high molecular weight (HMW) lignin derivatives possessing varied chemical properties were screened for their biological effects in order to obtain more information on the possible structural features of HMW lignin-related effects. The studied compounds were both commercial and in-house extracted lignin derivatives. Bioassays used include reverse electron transport (RET), Vibrio fischeri, Daphnia magna , and juvenile rainbow trout ( Oncorhynchus mykiss) hepatocytes. The studied lignin derivatives inhibited the in vitro systems and luminescence of V. fischeri bacteria to some extent–daphnids were not affected. Abstract The knowledge of chemical toxicity is necessary for risk assessment and ranking of chemicals according their hazard potential. The use of biopartitioning micellar chromatography has proven to be valid in predicting several biological activities of different kinds of compounds. In this paper, the relationships between retention of 66 organic pollutants and several ecotoxicity parameters are studied. Adequate correlations retention-ecotoxicity (LC 50 in fish and daphnia, EC 50 in green algae and daphnia, values of chronic toxicity in fish and green algae and bioconcentration factor in fish) of organic pollutants are obtained. Daphnia magna individuals were transplanted across 12 sites from three Spanish river basins (Llobregat, Ebro, Jucar) showing different sources of pollution. Gene transcription, feeding and biochemical responses in the field were assessed and compared with those obtained in re-constituted water treatments spiked with organic eluates obtained from water samples collected at the same locations and sampling periods. In order to predict the potential toxicity hazards of sediment remediation by dredging, an experimental laboratory simulation was made by investigating seven ratios of creosote-contaminated sediment (Lake Jamsanvesi, central Finland) and artificial lake water mixtures. Sediment was suspended in water at the ratios of 1:1, 1:2, 1:4, 1:8, 1:16, 1:32, 1:64, 1:128 v/v. The elutriates were analysed for the acute toxicity by photoluminescence bacterial and waterflea (Daphnia magna Straus) tests. The occurrence and sources of 21 perfluoroalkyl substances (PFASs: C4-C14, C16, C18 carboxylate, C4, C6-C8 and C10 sulfonates and C8 sulfonamide) were determined in water, sediment, and biota of the Llobregat River basin (NE Spain). Analytes were extracted by solid phase extraction (SPE) and determined by liquid chromatography triple quadrupole mass spectrometer (LC-QqQ-MS). As the number of legal and illegal gold mining sites increases in the Andes–Amazonia region, integrative methods to evaluate the effects of mining pollution on freshwater ecosystems are of paramount importance. Variability in δ13C values between individual Daphnia ephippia: Implications for palaeo-studies

The stable carbon isotope ratio (δ13C value) of Daphnia spp. resting egg shells (ephippia) provides information on past changes in Daphnia diet. Measurements are typically performed on samples of ≥20 ephippia, which obscures the range of values associated with individual ephippia. Using a recently developed laser ablation-based technique, we perform multiple δ13C analyses on individual ephippia, which show a high degree of reproducibility (standard deviations 0.1–0.5‰). The molecular masses of macromolecules and subunits of the extracellular hemoglobin from the fresh-water crustacean Daphnia magna were determined by analytical ultracentrifugation, multiangle laser light scattering and electrospray ionization mass spectrometry. The hemoglobins from hypoxia-incubated, hemoglobin-rich and normoxia-incubated, hemoglobin-poor Daphnia magna were analyzed separately. Abstract The purpose of this study was to determine the acute toxicity in aquatic organisms of one biocidal active substance and six metabolites derived from biocidal active substances and to assess the suitability of available QSAR models to predict the obtained values.
